| Barley is one of the oldest and most important cereal crops in the world.It has excellent resistance to drought,salt,low temperature and barren,and is an important model plant to improve the resistance of important crops.Stress associated protein(SAP)is a kind of zinc finger protein with A20/AN1 domains in stress response and stress regulation.This research analysis HvSAP gene family from the genome of barley and its physical and chemical properties,gene structure,the chromosomal location,phylogenetic tree.The expression pattern of HvSAP genes was analyzed by q RT-RCR.And the function of HvSAP6 gene was analyzed by gene cloning,subcellular localization and transgenic.The main research results are as follows:1.The HvSAP gene family members were identified using barley genome data.There were 13 HvSAP genes in barley.These 13 HvSAP genes coding sequence of length ranged from 360 bp to 885 bp and were located on the 1H,2H,3H,5H and 7H chromosomes of barley.Gene structure showed that 77% of the HvSAP genes had no intron and 15% had one intron,all of which contained the A20/AN1 zinc finger domain in barley.Expression analysis of 16 tissues of barley showed that HvSAP gene family members were expressed in all tissues except young inflorescences.2.The expression patterns of HvSAP genes under various abiotic stresses(salt,drought,low temperature,high temperature,and ABA)were analyzed by q RT-PCR,and it was found that most of the HvSAP genes could respond to these five stresses quickly,with different expression patterns,positive regulation and negative regulation.The results of subcellular localization showed that HvSAP6 and HvSAP12 proteins were expressed in the nucleus.3.The coding sequence of HvSAP6 gene was cloned and the overexpression vector of HvSAP6 was constructed.The vector was transferred into Arabidopsis thaliana by Agrobacterium-mediated method.The results showed that the root length,relative water content and proline content of the transgenic plants with HvSAP6 were significantly higher than those of WT under drought and salt stress,while the MDA content of the transgenic plants with HvSAP6 was significantly lower than that of WT.HvSAP6 can improve the stress tolerance of transgenic Arabidopsis thaliana. |
