In order to explore the effect of Diethyl aminoethyl hexanoate(DA-6)on potato growth and yield and quality,potential regulatory genes were explored,and the effects of DA-6 treatment on potato growth and development were explored from the physiological and molecular levels.In this study,potato buds,tissue culture seedlings,potted seedlings were used for the experiment.At the same time,by analyzing the transcriptome data of DA-6 processing in vitro bud germination,it was found that StWRKY40(by comparing the Arabidopsis homologous genes in the NCBI database was named WRKY40,so it was named StWRKY40 in this study),the gene expression level was significantly increased,and By successfully constructing the StWRKY40 gene expression vector,the growth curve was determined.The main results of this paper are as follows:(1)The results of 1 mg/L DA-6 treatment of in potato buds of Favorita and Rongziyu 5,showed that DA-6 treatment can promote the increase of the main root length and the number of roots of the two cultivars,and also promote the germination.(2)Fluorescence quantitative results showed that DA-6 treated potato buds and tissue culture seedling,StWRKY40 was up-regulated,gene cloning and bioinformatics analysis showed that StWRKY40 was 1083 bp in total length,StWRKY40 is a hydrophobic unstable protein,does not contain transmembrane structure,and contains typical WRKY functional domain.The construction of PET28a-StWRKY40 expression vector,transformation of E.coli,and spectrophotometric determination of OD600value showed The strain containing PET28a-StWRKY40 had a higher OD600 value under 10%sucrose and 5μmol ABA stress than the empty carrier alone.(3)DA-6 treatment promoting the root of Favorita tissue culture seedling,increasing the soluble protein and K+,Fe3+content of tissue culture seedling,Mg2+and Ca2+contents have no significant difference under the treatments of CK and DA-6,DA-6 treatment can reduce the content of hydrogen peroxide and increasing the peroxidase activity.(4)Using DA-6 to spray the leaves of the potato potted seedlings of Favorita and Rongziyu 5,the results showed that the treatment of DA-6 could promote the plant height and leaves of the pot seedlings of Favorita and Rongziyu 5,increase the chlorophyll content,increase photos synthesis rate of the two varieties,reduce the hydrogen peroxide content in the leaves of the two varieties,increase peroxidase activity,promote the accumulation of Mg2+and Fe3+,increase the number of tubers and the number of large and medium-sized departments.
