Our country is a big agricultural country.As one of the most important crops in our country,wheat requires a lot of water during its growth period.And our country is one of the countries with the most serious land desertification in the world.Drought has become one of the major factors restricting the growth of wheat and affecting wheat yield and quality.Therefore,finding genes involved in the regulation of drought resistance in wheat is important meaning for protecting food production security.In this study,hybrid wheat JM179 and its male parent BS7145 and female parent04YH27 were used as test materials,and their total chlorophyll content,free proline content,malondialdehyde content,superoxide dismutase SOD and soluble sugar content and other physiological indicators of drought resistance were used as test materials.Perform testing.The results showed that under drought conditions,the total chlorophyll content in wheat leaves decreased,but the material with stronger drought resistance had relatively higher total chlorophyll content,and its free proline content and superoxide dismutase SOD activity also increased significantly.high.In addition,drought treatment also led to an increase in malondialdehyde content and soluble sugar content in wheat.However,for materials with strong drought resistance,the increase rate of soluble sugar content was higher,but malondialdehyde increased less.Based on this,it can be inferred from the experimental data that the drought resistance of the three materials is ranked as follows: hybrid wheat seed JM179> male parent BS7145> female parent 04YH27.Glycosylation modification is an important post-translational modification of protein in plants,which participates in the regulation of various biological functions.Glycosylation modification methods are divided into two types: glycosylation and deglycosylation.Among glycosylases,glycosyltransferase(Glycosyltransferase,GT)is one of the most important enzymes,In deglycosylation enzymes class,no doubt,glycoside hydrolase(Glycoside hydrolase,GH)is one of the most important enzymes.In this study,we used CAZy database and hmmer software to search for and identify 211 glycosyltransferase family genes and 799 glycoside hydrolase family genes in wheat,Further bioinformatics analysis such as phylogenetic tree,gene structure,motif conserved domain,chromosome distribution,isoelectric point,etc.were carried out on the two families.An expression heat map was made by using the expression data of wheat GT family genes before and after drought treatment downloaded from the wheat expression website,and the genes with higher expression levels after drought treatment were screened out as the first part of candidate genes based on the results of the heat map.Then compare all the genes of the wheat GT family with the genes in the drought tolerance transcriptome database,and the result of the comparison is used as the second part of the candidate genes.The two candidate genes were integrated to obtain a total of 18 glycosyltransferase genes(TaGT186,TaGT204,TaGT157,TaGT138,TaGT195,TaGT169,TaGT32,TaGT145,TaGT66,TaGT77,TaGT92,TaGT140,TaGT155,TaGT167,TaGT175,TaGT178,TaGT130,TaGT187).Using the hybrid wheat JM179 and its male parent BS7145 and female parent 04YH27 before and after drought treatment as test materials,the Real time quantitative PCR experiment showed that the expression level of TaGT157 gene in the JM179 material under drought treatment was significantly higher than that of its parents.The gene may be involved in the molecular regulation of drought resistance of hybrid JM179.Building the subcellular localization vector GFP-TaGT157 of TaGT157 gene was constructed for subcellular localization experiments.The results showed that TaGT157 gene is expressed in the cytoplasm and cell membrane,and the previous subcellular localization prediction showed that TaGT157 gene may be localized in mitochondria It does not match the experimental results.It is speculated that the TaGT157 gene may not only participate in the regulation of drought resistance,but also has a wider range of functions to be sought.
