Microbial fertilizer can reconstructing soil healthy and preventing and controlling soil-borne diseases.However,the stability of field application effects is widely questioned.The user ignoring that the microorganism in the microbial fertilizer are in a dormant state.Generally,the product is simply dissolved in water and immediately applied to the soil.It is difficult to ensure the amount of microorganism and their active substance when applying to the soil.Result in the problems of high field application cost and unstable efficacy.Therefore,studying how to increase the effective amount of microorganism and active substance content in microbial fertilizer through field activation has great significance for correct application microbial fertilizerLvnonglin’ No.41 microbial fertilizer is a multifunctional microbial fertilizer manufactured by our laboratory,the Synergistic-Enhanced of antagonistic and growth-promoting ability),but there is still a lack of systematic research on the activation of Lvnonglin’ 41 under the field environmental conditions.Based on the good research foundation,we analyze the effects of amino acid water-soluble fertilizers on multiplication,secretion of active substances,and promoting plant growth and disease prevention of compound Bacillus,and better understand the activation conditions for applying Lvnonglin(?)41 in the field,provide theoretical guidance for the prevention and control of soil-borne diseases and scientific application methods of microbial fertilizer with field activation technology as the core and.The research results as follows1.By monitoring the living cell number,the inhibition zone and the content of IAA,it was verified that the nutritional carrier amino acid water-soluble fertilizer of Lvnonglin(?)41 can activate compound bacillus multiplication and secretion of active substances under field conditions.The results showed that the amount of living cell of Lvnonglin(?)41 which diluted 20 times with clean water without activated was only 2.60×108 cfu/mL,and the diameter of inhibition zone was only 0.92 cm,and the IAA was only 54.3 ug/mL.Lvnonglin(?)41 is diluted 20 times with clean water and placed at room temperature(24-35℃)with ventilated by oxygen pump for 24 hours.The living cell number in the activated Lvnonglin(?)41 solution is 3.85×109 cfu/mL,the diameter of the inhibition zone was 1.57 cm.The yield of IAA was 98.5 ug/mL.The living cell number,diameter of the inhibition zone,and the yield of IAA of activated Lvnonglin(?)41 was 13.8 times,70.7%,81.4%higher than that of the unactivated Lvnonglin(?)41.2.Through single factor and response surface optimization experiments,confirmed the field activation conditions for Lvnonglin(?)41 were:dilution 20-50 times,temperature 25-37℃,and ventilation time 36 h by comprehensive analysis.The diameter of inhibition zone was 1.41-1.86 cm,and the yield of IAA was 86.0-118.9 ug/mL.3.Through the pot experiment evaluated the effect of the activated Lvnonglin(?)41 on preventing and controlling soil-borne diseases.The results showed that compared with the Lvnonglin(?)41 treatment diluted to 500 times without activation,the Lvnonglin(?)41 treatment diluted to 500 times after activation had a significant effect on disease resistance and growth promotion to banana seedlings,and the specific performance was on height,stem thickness,fresh weight above ground and fresh weight below ground increased by 14.8%,26.4%,9.4%,and 1.7%respectively;the number of fungi,actinomycetes and bacteria in rhizosphere soil increased 45.5%,23.7%,and 5.3%respectively;The amount of Foc in the rhizosphere soil was reduced 15.4%;the disease incidence of banana wilt was reduced to 25.0%,and the control treatment was increased to 46.72%.4.The pot experiment evaluated the effect of Lvnonglin(?)41 on tomato fusarium wilt after field activation.It was found that compared with the treatment of Lvnonglin(?)No.41 which was diluted to 500 times without activation,the treatment of Lvnonglin(?)No.41 which was diluted to 500 times after activation had significant effects on promoting tomato growing and preventing diseases.The height,stem thickness,Leaf area,fresh weight,and root length increased by 27.1%,27.2%,21.5%,25.4%,and 17.1%respectively;defense enzymes such as PAL,PPO,POD,and CAT increased by 20.0%,14.0%,22.2%,13.2%,respectively,the disease incidence of tomato fusarium wilt was reduced to 22.2%,and the control effect was increased to 66.1%5.Field experiments were conducted to evaluate the effects of activated Lvnonglin(?)41 on preventing and controlling tomato fusarium wilt and rhizosphere soil nutrients.The results showed that compared with the treatment of Lvnonglin(?)No 41 which was diluted to 500 times without activation,the treatment of Lvnonglin(?)No 41 which was diluted to 500 times after activation had the most significant function of promoting tomato growing and preventing diseases.The height,stem thickness,leaf area,and chlorophyll content increased 16.9%,20.8%,27.6%,and 4.4%,respectively The soluble sugar content,soluble protein content,lycopene,and Vc content of tomatoes increased 10.8%,13.5%,15.7%,15.6%,respectively.The soil organic matter,ammoniacal nitrogen,available phosphorus and available potassium content increased 3.2%-4.5%、4.7%-6.8%、1.5%-2.4%、2.9%-7.1%,respectively.The results of GC-MS analysis of metabolomics showed that,the contents of Ethanedioic acid,L-Proline,Glutamine and Unidentified substance(Unknown9)showed regular changes in different treatments.The tomato fusarium wilt disease incidence of tamato which treated with the activated Lvnonglin(?)41 was reduced to 20.0%,and the control effect was 74.3%;while the disease incidence of the unactivated Lvnonglin(?)41 treatment was 40.0%,and the control effect was only 36.9%In short,The activated Lvnonglin(?)41 compound microbial fertilizer has the functions of significantly increasing yield and quality,inducing diseases resistance and preventing diseases. |
