In recent years,the anti-photoaging activity of royal jelly has attracted more and more attention,and the series of skin care products such as royal jelly facial masks are also very popular.However,the proteins in royal jelly have the disadvantages of causing allergic reaction,unstable storage and not easy to be absorbed by human skin.In order to solve the above problem and make royal jelly’s anti-photoaging effect better play.We used proteolysis technology to hydrolyze royal jelly.The physicochemical properties of lyophilized powder of the royal jelly,major royal jelly proteins(MRJPs),enzymatic hydrolysates of royal jelly and enzymatic hydrolysates of MRJPs were characterized.The anti-photoaging activity of the four kinds of samples and 10-HDA were been tested through the human fibroblasts photoaging model,and their anti-photoaging mechanism were discussed.The results are as follows.1.The main protein molecular weight of royal jelly ranged from 40 kDa to 70 kDa,and the peptide molecular weight of enzymatic hydrolysate ranged from 180Da to 1500Da.The distribution of amino acids in the four samples was consistent.Aspartic acid has the highest content,accounting for about 20%of the total amino acids.The results of FTIR showed that the 1648 cm-1 peaks of royal jelly and MRJPs shifted to 1654 cm-1 and 1652 cm-1 after enzymolysis,which means the amide band of protein was obviously broken by enzymatic hydrolysis.2.854 enzymatic hydrolysates of royal jelly and 426 enzymatic hydrolysates of MRJPs were obtained by mass spectrometry test,and 306 royal jelly hydrolysates peptides and 108 MRJPs hydrolysates peptides with antioxidant activity were screened.3.We established the photoaging model of human skin fibroblasts irradiated with 365 nm UVA at a irradiation dose of 10 J/cm2.In this model,The concentration of royal jelly,MRJPs,enzymatic hydrolysates of royal jelly and enzymatic hydrolysates of MRJPs were 100 μg/mL and 10-HDA was 5 μg/mL.The samples downregulated MMP1 in the gene expression of 21.74%,36.94%,34.53%,50.5%and 62.78%respectively.And the samples increased COL1α1 protein content of 8.95%,15.01%,15.08%,18.94%and 43.58%respectively.Five kinds of samples exerted anti-photoaging activity by inhibition of MMP1 gene expression and COL1α1 protein synthesis.The results showed that Enzymatic treatment can improve the anti-photoaging activity of royal jelly and MRJPs.
