At present,wheat hybrid production using two-line hybrid technology has been industrialized.Photo-Thermosensitive Genic Male Sterile Lines are the core materials for two-line hybrid system,and its fertility is regulated by light and temperature.Although previous researchers have done a lot of research on fertility conversion mechanism,but it is not clear.LncRNA is abundant in plants,playing important roles in plant growth and development,biological and non-biological stress response by regulating gene expression.Our previous studies have cleared that anther fertility of photo-thermo sensitive Genic Male Sterile Wheat BS366 could be affected by environments.In this study,long non cording RNAs were identified from anthers in different light-temperature by deep sequencing.To well know the function of lncRNAs,GO annotation and KEGG pathway were performed on targets of lncRNA.Moreover,it was found that the differential expression of lncRNA27195 and its targets TaRTS was involved in pollen development.Thus,gene and protein structure of TaRTS were further analyzed.In addition,gene expression of lncRNA27195 and its targets TaRTS in different tissues and different treatments were also analyzed.The results are as fellows:(1)In total 663 differential expressed lncRNA were obtained,which 249 were up regulated and369 were down-regulated.(2)Enrichment analysis for targets of lncRNAs showed that: these genes were enriched on regulation of transcription DNA-templated,vacuolar membrane and nucleic acid binding in environment of low temperature long and short photoperiod.The genes were enriched on multicellular organism developmen,chloroplas in environment of high temperature long and short photoperiod.(3)LncRNA27195 and TaRTS which involved in pollen fertility were screened by transcriptome sequencing.The results showed that lncRNA2719 was secretory protein with α-helix structure and was hydrophobic.(4)The expression level of lncRNA27195 and TaRTS are higher in stamen of wheat than in other tissues.lncRNA27195 and TaRTS are specifically expressed in stamens,which are mainly involved in the regulation of anther development.(5)Light and temperature had significant effects on the expression of lncRNA27195 and TaRTS in wheat.High temperature and long photoperiod can promote the expression of lncRNA27195 and TaRTS,fertility was restored.(6)Proper amount of MeJA could promote the expression of lncRNA27195 and TaRTS,while high concentration of MeJA inhibit expression;SA can inhibit the function of MeJA and then the expression of lncRNA27195 and TaRTS were.inhibited.In conclusion,lncRNA plays important roles in temperature and light-induced fertility transformation of PTGMS wheat.Light-induced male sterility may be mainly due to the abnormal expression of genes in sucrose and other energy anabolic pathways.Temperature-induced male sterility may be mainly caused by gene blocking or slowing down in the process of energy transport.In addition,by bioinformatics analysis and expression level analysis on lncRNA27195 and TaRTS,it was further proved that lncRNA and its target gene could regulate pollen fertility and are regulated by MeJA.This gene may be secreted into pollen along with tapetum apoptosis,providing essential nutrition for pollen wall formation and pollen growth and development.
