Ramie is one of the important economic crops in south of China,It is full of natural fiber and other substances.Textile fiber quality depends on its value,the fiber development has important significance for cultivating high quality in fiber and ramie varieties.This study had performed a long non-codingRNA(lncRNA)sequenced by using the stem bark tissues collected from Zhongzhu No.1,Identified mRNA and lncRNAs may be associated with ramie fiber development,Bioinformatics was used to annotate its functions and predict its targeting mode.On this basis,the interaction and biological function of lncRNA with its target genes were identified.this study detected lncRNAs in ramie,and our results provide important basis for understanding the mechanism of fiber development,which provided an important theoretical basis for the formation of ramie bast fiber.Progress has been made as follows:(1)The TPS and MPS of Zhongzhu No.1 were used to construct a library for transcriptome sequencing.A total of 4316 lncRNAs were identified and 1287 lncRNAs were differentially expressed.Among the 4520 PCTs,75 were identified as homologous genes of Arabidopsis SCW biosynthesis genes.In which 6 mRNAs was predicted were targeted by 10 lncRNAs.Bioinformatics predicted that these 6target genes were homologous to Arabidopsis fiber development related protein MYB,NAC,IRX15 L,LBD,and BLH1,and they were speculated to be related to fiber development,while 10 lncRNAs mainly regulated the function of target genes through targeting.It is speculated that they are related to the development of ramie fiber.(2)The candidate gene LTCONS00034183 and its target gene wholeGLEAN10025325 were cloned by PCR using Zhongzhu No.1 phloast fiber c DNA as template.The transgenic plant was obtained by constructing the overexpression vector and flocculating the plant with blob infection.The section of wholeGLEAN10025325 transgenic plant was observed,and it was found that the overexpression of this gene in Arabidopsis increased the content of xylem fibro cells and The phloem fibers increased significantly compared with that of wild type Arabidopsis at the same period.(3)Co-transformed transgenic Arabidopsis thaliana with LTCONS00034183 and its target gene wholeGLEAN10025325 showed that the xylem fiber content of the plant was decreased and the fiber content was slightly decreased compared with that of the wholeGLEAN10025325 transgenic Arabidopsis at the same period.It is suspected that the downregulation of LTCONS00034183 on target gene wholeGLEAN10025325 affects fiber development.This study lays a foundation for the biological functions of ramie lncRNA and target genes on fiber development,which is of great significance for broadening and enriching the understanding of the functions and mechanisms of plant lncRNA and fully revealing the molecular mechanism of ramie fiber development.Meanwhile,it also provides effective candidate genes for ramie high-quality fiber breeding.
