| Ramie is one of important textile materials in our country, and how to increase its yields and improve its qualities are getting more and more attention. The correlation between fiber quality, chemical components and key enzymes in different ramie varieties was been found by measuring the activity of the key enzymes (β-glucan enzyme and sucrose synthetase), quality indicators (single fiber size, breaking strength). Then the best favorable conditions of β-glucan enzyme activity was determined.The main results are as follows:1. The best suitable condition for measuring the activity of β-glucan enzyme was incubation at50℃about30min and then on the boiling water bath for10min.2. The DNS method showed that the activity of β-glucan enzyme was found to changed along with the growth of plants. The total variation into rising-down-up-dropped four stages. Before the grow date20days, the activity of beta glucan enzyme was stronger and high activity period is longer in XiangZhu No.3and Ya ma in Yizhang than other three ramie varieties, after then the two was lower. To the other side, before the date20days, the β-glucan enzyme activity of XiangZhu No.1, Empty bar ma in Xiangtan and Plexus hill ma in Pingjiang was low,after then continued to rise last remained relatively stable.Spectrophotometric method showed that sucrose synthase activity of ramie fiber was also changed with plant growth. The overall trend is divided into down-up-down three phases. The activity of sucrose synthase was a rising trend in XiangZhu No.1and XiangZhu No.3during grow date15to20.The change of sucrose synthase activity in Empty bar ma in Xiangtan and Ya ma in Yizhang was larger than other three high production varieties, and high activity period is shorter.3. Cellulose kept a rising trend with plant growth, but the content of pectic substances and hemicellulose in the ramie fiber is a downward trend, different varieties has different changes.The change rule of single fiber length, diameter, length to diameter ratio and single fiber breaking strength was increase gradually along with the growth and development of fiber. Length to diameter ratio represent fiber count. The high-quality varieties has bigger single fiber size because length to diameter ratio of Plexus hill ma in Pingjiang, XiangZhu No.3and XiangZhu No.1were lager than others in grow date of45. The breaking strength in XiangZhu No.3and Ya ma in Yizhang was the strongest. 4. The β-glucan enzyme activity was more better in high quality varieties XiangZhu No.3and Ya ma in Yizhang, and the period to maintain high activity was longer. And β-glucan enzyme activity is low in Low-quality varieties before the early stage of the development, the longer period to maintain high enzyme activity was after then. The β-glucan enzyme activity was positively related to single fiber breaking strength, but pectic substances and hemicellulose was the opposite.And sucrose synthase activity in low-yielding varieties Empty bar ma in Xiangtan and Ya ma in Yizhang changed greatly in the early development, the period to maintain high is shorter in the late development, the high production was opposite. Sucrose synthase activity may has positive correlation to cellulose content and length to diameter ratio. |
