Identification Of Cordyceps Militaris Strains And Optimization Of Cordycepin Extraction Process And Product Development

Cordyceps militaris,also known as the northern Cordyceps sinensis,has been considered as an alternative to Cordyceps sinensis in recent years.Cordycepin is one of the biologically active substances,which has the function to reduce blood lipids and anti tumor and thrombosis.Although large-scale production is achieved of C.militaris in China,the application is still relatively weak.Therefore,there are bright prospects for improving the extraction yield,optimizing the extraction process and developing the processing technique.In this paper,we select excellent strains through the separation of C.militaris tissue from wild,optimizes the ultrasonic water insectin process and obtains the best extract concentration through orthogonal experiment,and preliminary develop the processing technology of Cordyceps tea and Chinese caterpillar fungus medicine.The results are as follows:1.Isolation and domestication of C.militarisSeven strains of C.militaris were isolated from wild strains and identified by morphology(colony observation and light electron microscope observation)and molecular identification by building a developmental evolutionary tree.Seven strains are very homologous.According to the results of phylogenetic analysis,the isolated strain belongs to C.militaris.Among them,The growth rate of YC-C strain was faste,after 15 generations continuous,the change of cordycepin content was not significant,which indicated that the genetic stability of YC-C strain is good.2.Optimization of Cordycepin extraction processThe effect of ultrasonic water extraction of Cordycepin and adenosine was investigated by orthogonal experiments in three fators including time,temperature and the ratio of material to liquid.The experimental results show that at the extraction temperature is 50°C,the ratio of material to liquid is 1/40 and the ultrasonic extraction of 1.5 h,the extraction amount of Cordycepin reached the highest.YC-C strains is the maximum value of herb extract in seven strains which reaches 5.6794 mg/g.3.Cordyceps bag tea and Cordyceps granulesBased on odor,color and taste,the preliminary formula of Cordyceps bag tea is determined by single factor experiment as 1 g Cordyceps militaris,0.4 g red dates,1cinnamon round,2 grains of wolfberry,0.1 g tangerine peel and one rose.Different Cordyceps granules are preliminarily screened out for different tastes,lemon black tea flavor: 0.7 g cordyceps powder,1.4 g maltodextrin,0.7 g black tea powder,0.04 g citric acid,0.02 g Ansai honey;cranberry flavor: 0.7 g cordyceps powder,1.4 g maltodextrin and0.7 g cranberry flour;passionfruit flavor: 0.7 g cordyceps powder,1.4 g passionfruit powder,0.5 g seabuckthorn powder and 0.02 g Ansai honey.And the test result indicated that the three flavor formulations of Cordycepin is little different from that in the market products.
