Inherently disordered proteins(Intrinsically disordered proteins,IDPs)are a class of proteins that are not folded as a whole or locally in a natural state and lack stable three-dimensional structures but are able to perform biological functions normally.They are ubiquitous in the proteome and participate in a series of biological processes such as signal transduction,transcriptional regulation,stress response,and so on.At present,there are few reports on the function of woody plant IDPs in salt stress response.In this study,the gene PtrIDP1(Potri.010G161200.1)was cloned from Populus trichocarpa(Torr.and Gray)and its protein sequence,gene expression and function were studied.(1)The CDs sequence length of the PtrIDP1 gene is 423bp,and its encoded protein contains 140 amino acids.The PtrIDP1 gene is a stable and hydrophilic protein,and its isoelectric point(pI)is 5.58,the lipolysis index is 58.71,The total average hydrophilic coefficient is-0.716,the instability coefficient is 29.60,indicating thatAccording to the systematic evolution tree,the PtrIDP1 protein is closely related to the PeIDP1 from P.trichocarpa.(2)Real-time PCR analyses showed that PtrIDP1 gene could be expressd in ifferent organs of P.trichocarpa,PtrIDP1 gene was expressed in roots,stems and leaves of P.trichocarpa,among which the lowest expression was found in roots and the higher relative expression was found in stems and leaves.The expression of PtrIDP1 genes under drought and salt stress was analyzed,which showed that salt stress promoted PtrIDP1 expression and PEG could regulate PtrIDP1 expression.(3)Construction of subcellular localization expression vector,transient transformation of onion epidermal cells,observed under laser confocal microscope,PtrIDP1 gene-encoded proteins located in the nucleus.(4)Cloning and constructing pROK2-PtrIDP1 plant overexpression vector,using Agrobacterium tumefaciens mediated transformation of gene into and analyzing the function of gene in salt stress response.Under salt stress,the plant height of transgenic plants was significantly higher than that of non-transgenic plants.Compared with non-transgenic plants,the contents of chlorophyll、SOD、POD transgenic plants increased significantly,while the content of MDA decreased.Above physiological indexes showed that the overexpression of gene in poplar significantly improved salt tolerance of transgenic plants.to sum up,genes play an important role in P.trichocarpa salt stress response.
