Histone acetylation is an important epigenetic modification and has important regulationary functions in chromatin structure change and remodeling, gene expression and multiple cellular processes. Histone acetylation is regulated by histone acetyltransferase (histone acetyl transferase, HAT) and histone deacetylases (histone deacetylase, HDAC), respectively.HDACs are involved in growth, development and stress response in plants. Populus HDT902 is amember of HD2 family, and, in this study, the results of its expression and functions were analyzed.The Open Reading Fram (ORF) was 840bp in length of PtHDT902 gene was cloned from Populus trichocarpa..The protein sequence of PtHDT902 gene encoding cDNA by bioinformatics analysis showed that, HDT902 protein has a typical zinc-finger domains. HDT902 protein has a high identity in amino acid sequence with peach PpHDT5801 and is closely related to PpHDT5801 in evolution. Promoter analysis showed that the promoter sequence of PtHDT902 gene contains multiple cis-acting elements including Box 4, Box I, GARE-motif, ERE, HSE, LTR and a plurality of stress-related elements. Subcellular localization analysis showed that PtHDT902 protein was localized in the nucleus. PtHDT902 gene expression in various organs and in response to cold and salt stresses were analyzed by Real-time PCR results showed that PtHDT902 was expressed in root, stem and leaf. The expression of PtHDT902 was regulated by cold and salt stress, indicating that HDT902 gene was involved in cold and salt stress responses. To study the functions of PtHDT902, a plant expression vector was constructed and transformed into Arabidopsis and 84K poplar by Agrobacterium-mediated method,the expression of PtHDT902 in Arabidopsis promoted the growth of root, and the expression of GA synthase genes including AtKO1, AtKAO2, AtGA20ox1, AtGA20ox2, AtGA20ox4 and AtGA3oxl was up-regulated. In addition, the expression of PtHDT902 gene 84K populous promoted callus induction from leaves.
