Since the discovery of penicillin in 1928,antibiotics have become the gospel of human and animal health.Due to the abuse of antibiotics,antibiotic resistance genes continue to accumulate and spread.Antibiotic resistance genes have now been listed as a new type of environmental pollutant,bacterial Drug resistance has become a global public health threat and has received widespread attention.Escherichia coli can cause diarrhea,hemorrhagic colitis,dysentery,hemolytic uremic syndrome and other diseases,and bring huge economic losses to farmers in the process of livestock and poultry breeding.However,in recent years,the resistance rate of Escherichia coli to antibiotics has gradually increased,which has brought huge economic losses,but also brought huge threats to human health.In order to understand the prevalence of resistance genes in sheep manure and soil in the southern foot of Tianshan Mountains,Xinjiang,as well as the resistance of sheep manure Escherichia coli and soil Escherichia coli and the resistance genes carried by them in different locations,this study was collected from the southern foot of Tianshan Mountain.Korla went to 10 locations in Kashgar in the southern foot of the Tianshan Mountain sheep manure and soil samples for drug resistance gene detection,and isolated Escherichia coli,the isolates were subjected to quinolones,aminoglycosides,tetracyclines,sulfonamides,β-lactams and viscose Drug susceptibility test of antibiotics such as bacteriocins and detection of drug resis tance genes.(1)Detection of resistance genes in manure and soil samplesThe PCR method was used to detect quinolones,aminoglycosides,β-lactams,tetracyclines,sulfonamides and colistin-related resistance genes in 50 sheep manure samples and 50 soil sa mples collected from 10 locations.The results showed that sul1、sul2、tet A、aad B、bla CTX-M、gyr A in manure and soil samples were detected in 10 locations.tet B、aad A1、aph(3’)-I、pro P-2、amp G、bla CIT、bla TEM、mcr-1 are different depending on the location.(2)Isolation and identification of manure and soil Escherichia coliStrain isolation of Escherichia coli in manure and soil was carried out by ordinary agar medium,beef extract peptone medium,Mac Conkey medium and eosin methylene blue medium.After Gram staining,biochemical identification,molecular biology identification and Qualcomm The quantitative microbial phenotype identification analysis system identified 148 Escherichia coli.(3)Detection of Escherichia coli drug resistance and drug resistance genesThe drug-sensitive paper method was used to analyze the resistance of 11 kinds of antibiotics to Escherichia coli isolates,and the resistance genes carried by them were detected by PCR method.The results showed that the drug resistance rate of sulfisoxazole was generally high,with triple drug resistance and quadruple drug resistance being the main ones,and five d rug resistance less,but all strains were resistant to ciprofloxacin and enrofloxacin and the rates are all zero.The types of resistance genes detected in Escherichia coli were less than those detected in manure and soil samples.sul1、sul2、tet A、aad B、bla CTX-M were detected in 10 locations,while tet B、bla CIT、bla TEM were not detected.aad A1、aph(3’)-I、pro P-2、amp G、mcr-1 are different due to different locations,and the detection rate of sul1、sul2、tet A、aad B、bla CTX-M is compared with tet B、aad A1、aph(3’)-I、pro P-2、amp G、bla CIT、bla TEM、mcr-1 are relatively high,and they are the main resistance genes that are prevalent in Escherichia coli.The results of multilocus sequence typing showed that the prevalence of Escherichia coli in 10 locations differed in different locations.ST10,ST86 and ST155 were prevalent in many places,and other ST types also had a certain prevalence.Through this study,we can understand the prevalence of drug resistance genes in sheep manure and soil in the southern foot of Tianshan Mountains,X injiang,as well as the drug resistance of Escherichia coli and the prevalence of drug resistance genes carried in 10 locations,so as to provide references for veterinary clinical rational use of drugs,providing a basis for preventing the further spread of drug resistance.
