Raspberry is a deciduous shrub belonging to the genus Rubus of Rosaceae.The fruits,leaves and seeds of raspberry are rich in various nutrients and have high value in medical and edible aspects.China is rich in wild resources of the genus Rubus.Collecting local wild resources,domestic and foreign species resources,understanding its genetic diversity,and making correct evaluations are important prerequisites for carrying out breeding work.Molecular marker technique has been widely used to analyae the genetic diversity of crop resources,however,the application of SRAP for Rubus has not been reported yet.Therefore,in this study,we collected the wild raspberry resources in northern China,domestic cultivated,and some wild resources in Guizhou Province,using SSR and SRAP molecular marker techniques,and clustered analysis to analyse the genetic diversity and kinship of the collected resources.The main purpose was to provide more resource information for raspberry breeding,so as to improve the efficiency of breeding.The main results were as follows:(1)In this study,a raspberry SRAP reaction system was constructed for the first time,and 10 pairs of primers that could be used for raspberry SRAP research were screened out,including Me1/Em2,Me1/ Em4,Me2/ Em2,Me2/Em4,Me3/Em1,Me3/Em4,Me4/Em1,Me4/ Em4,Me3/Em3.(2)Ten pairs of SSR primers were used to amplify 76 samples,and the number of bands amplified in each sample ranged from 13 to 29;Ten pairs of SSR primers were used to amplify 76 samples,and the number of bands amplified in each sample ranged from 12 to 36.(3)Based on the results of SSR and SRAP markers,the genetic diversity of wild resource populations in Guizhou(A1),cultivated varieties(A2)and wild resources in the northeast of China(A3)were analysed.It was found that the ratio of polymorphic loci,effective alleles,Nei’s index and Shannon I index of the A2 population are higher than those of the A1 and A3 populations.(4)Analysis of among 76 raspbery germplasm resources based on SSR markers,among which the genetic similarity coefficient was between 0.473 to 0.982.Based on the results of cluster analysis,these samples were divided into three categories,and in the second category,they were divided into there subcategories.Moreover,the genetic similarity coefficient among these materials ranged from0.520 to 0.972 analysed by SRAP markers,according to the result of cluster analysis,these samples were divided into two categories,and in the second category,there were five subcategories.By the two molecular marker technologies,it was finally possible to separate cultivated varieties from wild resources,and separate wild resources in Northeast China from wild resources in Guizhou.
