Rice is an important food crop in China.Rice blast is one of the important diseases of rice,which is caused by(Magnaporthe oryzae syn.Pyricularia oryzae).It is an important agricultural pathogenic microorganism and causes huge yield loss in rice production.MoCrbp1 and MoCrg1 are two proteins with unknown functions in M.oryzae.Transcriptome sequencing revealed that the expression levels of these two genes in the deletion mutant of MoCre A,a carbon metabolism repressor of M.oryzae,were significantly reduced.Protein domain analysis found that MoCrbp1 contains the BBTB/POZ domain;while MoCrg1 contains the Zn2Cys6motif unique to fungi and is homologous to GAL4-like transcription factors.The two genes were knocked out by the strategy of protoplast transformation through homologous recombination.When compared with the wild type strain Guy11,the gene deletion mutantsΔMoCrbp1 andΔMoCrg1 showed less pathogenicity in both barly and rice plants.And the number of conidia produced in the mutants was reduced.Both mutants were more sensitive to hydrogen peroxide.The appressorium formation ofΔMoCrbp1 mutant conidia was prolonged.The research results laid the foundation for further exploration of the biological function and pathogenic mechanism of MoCrbp1 and MoCrg1 in M.oryzae.
