Walnut(Juglans regia)because of its high utilization value,growing area,which leads to the by-product of walnut yield is also increased.Walnut shell is randomly discarded in the field or simply processed into activated carbon and filter material for use;diaphragma juglandis is directly used as traditional Chinese medicine,but due to the low utilization rate,resource waste and envionmental pollution are caused.In this study,walnut shell and diaphragma juglandis were used as raw materials,and a variety of organic solvents were used for high efficient extraction.FT-IR,GC/MS and UPLC-QTOF-MS/MS were used to analyze the chemical components,and the composition,function and anti-cancer potential of the bioactive components in diaphragma juglandis and walnut shell were explored,which laid the foundation for the value-added enhancement and multi-level utilization of the by-product of walnut.The results are as follows:(1).Efficient extraction of walnut shell and diaphragma juglandis with various solvents:It was found that there were esters,ketones and acids in walnut shell by FT-IR.There are alcohols,esters and ketones in diaphragma juglandis.Walnut shell and diaphragma juglandis were extracted efficiently by ethanol,acetone and ethyl acetate.(2).Analysis of volatile components and functions of walnut shell and diaphragma juglandis:The results of GC-MS analysis showed that there were 175 kinds of volatile substances in walnut shell,including myristic acid,trans oleic acid,furfuran acid and verbenol.For the first time,12 volatile compounds were detected in walnut shell,such as naringin and ionone.There are 186 kinds of volatile substances in distracted wood.The main volatile substances are D-gamma-tocopherol,cis-10-heptadecaenoic acid,s-(7-octenyl)sodium bisulfate and monoethyl succinate.For the first time,14 kinds of volatile substances were detected in diaphragma juglandis,such as Sakura glycoside and ionone.There are 21 kinds of substances in walnut shell and diaphragma juglandis,the main components are trans oleic acid,cherry blossom glycoside and astaxanthin,among which 5 kinds are new substances.The extraction effect of ethyl acetate solution from walnut shell is the best,and the extract is suitable for biomedical and chemical raw materials.The extraction effect of ethanol solution of diaphragma juglandis is the best,and the extract plays an important role in medicine and chemical industry,for example,heptanic acid can be used to synthesize antifungal drugs;naringin has obvious anti-inflammatory effect.(3).Analysis on the characteristics and uses of nonvolatile components of walnut shell and diaphragma juglandis:867 kinds of nonvolatile substances were found in walnut shell by UPLC-QTOF-MS/MS,mainly including dimethyl adipate,styrene and triphenyl phosphate.Among them,28 kinds of new substances were found in walnut shell for the first time,such as sebacic acid,5,8,12-trihydroxy-9-octadecanoic acid and tocopherol acid.There are 1019 kinds of nonvolatile substances in diaphragma juglandis,mainly including epicatechin gallate,hypaconitine and kaempferol,among which 45 are newly found.There are 15 kinds of nonvolatile substances found for the first time in walnut shell and diaphragma juglandis,such as epicatechin gallate,sphingomyelin and sodium phenolate.The extraction of ethyl acetate from walnut shell is the best.Non volatile substances are suitable for the development of biological medicine and flavor and fragrance products.The ethanol extract of diaphragma juglandis has the best extraction effect,and the nonvolatile substances are suitable for chemical and pharmaceutical raw materials.(4).The main potential anticancer factors of walnut shell and diaphragma juglandis:There are 15 potential anticancer active factors in walnut shell and diaphragma juglandis,among which epicatechin gallate,kaempferol and furane contribute nearly 10%of the anticancer effect,and there are unknown functional substances around these substances Such as heptanoyl carnitine in anti-inflammatory,antibacterial and other aspects of research potential.In this paper.walnut shell and diaphragma juglandis are taken as the research object,and the advanced FT-IR,GC/MS and UPLC-QTOF/MS are used to systematically analyze the component characteristics and uses of volatile substances and nonvolatile substances in walnut shell and diaphragma juglandis.The potential anti-cancer factors of nonvolatile substances in walnut shell and distracted wood were systematically analyzed by principal component analysis,which laid a foundation for multi-level utilization of walnut shell and distracted wood and for improving the added value of walnut.
