| Wool production traits are the most important economic traits of wool rabbits.Finding SNP markers related to hair production of long wool rabbits is the basic work to improve wool production traits.In this study,breeding populations of Yin meng wool rabbits were used as test materials.FGF5、KRT26、FAIM、PROP1、HOXA4 and FZD3 genes were selected as candidate genes.Polymorphisms in their 5 ’、3’ regulatory regions and coding regions were determined using mixed pool sequencing.A total of 21 SNP loci were found,and SNPs were typed using MALDI-TOF technology.The wool production traits of rabbits,gross wool rate,hair length,wool fiber diameter,and wool fiber diameter were measured.SAS8.0 statistical software analyzed associations of candidate gene 5 ’,3’ regulatory regions and coding regions with SNP loci and hair production traits.The main findings are as follows:(1)In this experiment,it was found that two loci had a significant effect on hair production in long wool rabbits(P<0.05).The effect mode of FGF5 loci 705 on hair production was additive effect and dominant effect,and the effect mode of HOXA4 loci 649 on hair production was dominant effect.One locus had a significant effect on hair production in long wool rabbits(P<0.01),and the effect mode of KRT26 gene locus 1065 on hair production was additive effect.(2)In this experiment,a total of 2 sites were found to have an effect on the villi length of long wool rabbits.FAIM gene locus 260 had a significant effect on the villi length(P<0.05),and its mode of action was additive effect.The effect of KRT26 gene site 1065 on villus length was significant(P<0.05).(3)In this experiment,a total of 7 sites were found to have effects on the diameter of long-hair rabbit hair fiber,among which sites 649 of HOXA4 gene,693 of KRT26 gene and1064 of KRT26 gene had significant effects on the diameter of villi fiber(P<0.05).FZD3 gene locus 750 and KRT26 gene locus 345 and 929 had significant effects on villi fiber diameter(P<0.01),PROP1 gene locus 410,KRT26 gene locus 345,639 and 1064 had significant effects on coarse hair fiber diameter(P<0.05).The effect of 750 locus of FZD3 gene,649 locus of HOXA4 gene and 1064 locus of KRT26 on the diameter of villi fiber was dominant.The effect of KRT26 345,693,and 929 on the diameter of villi fiber was characterized by dominant effect and additive effect.The effects of PROP1 loci 410,KRT26 loci 345,693,and 1064 on the diameter of coarse wool fiber are shown as dominant effect and additive effect.(4)In this experiment,a total of 5 loci were found to have an effect on the hair rate of long-haired rabbits.FGF5 loci 705,PROP1 loci 410,FZD3 loci 750,KRT26 loci 736 and 929 had significant effects on the hair rate(P<0.05).The effect of KRT26 loci 736,929 and FGF5 loci 705 on gross hair was additive.The effect of PROP1 locus 410 on the gross hair rate was dominant.The effect of FZD3 locus 750 on the gross hair rate was shown as dominant effect and additive effect.(5)In this experiment,six loci were found to have an effect on the coefficient of variation of wool fiber diameter,among which,locus 345,736 and 833 of KRT26 gene had a significant effect on the coefficient of variation of wool fiber diameter(P<0.05);locus 750 of FZD3 gene had a significant effect on the coefficient of variation of wool fiber diameter(P<0.01).There was a significant effect of site 260 of FAIM(P<0.01)and site 649 of HOXA4(P<0.05).Locus 345,736,833 of KRT26 The mode of action of FZD3 gene 750 on the coefficient of variation of wool fiber diameter is dominant effect;the mode of action of HOXA4 gene 649 on the coefficient of variation of wool fiber diameter is additive effect;the mode of action of FAIM gene 260 on the coefficient of variation of wool fiber diameter is additive effect and obvious effect Sexual effects.(6)The results of haplotype construction of SNP locus of the target gene and the analysis of related hairy characters and effects showed that: there were significant differences between six haplotypes of KRT26 gene(P<0.05).In conclusion,12 loci were found to have a significant effect on the hairy traits of Angora rabbits.Haplotypes and haplotypes interaction in KRT26 gene had a significant effect on the hairy traits of Angora rabbits.The results showed that FGF5、KRT26、FAIM、PROP1、HOXA4 and FZD3 genes may be candidate genes affecting the wool production traits of long wool rabbits..The 12 SNP loci screened in this study are expected to be used as molecular markers for the breeding of wool producing traits in Angora rabbits. |
