| The polysaccharide component in traditional Chinese medicine can be used as an immunopotentiator or adjuvant to enhance the immune function of the animal body,thereby playing an immune enhancement role,and has received extensive attention in recent years.This study mainly developed a traditional Chinese medicine polysaccharide,which was used to enhance the vaccine immune effect.Through different extraction methods,the polysaccharide extraction rate of four Chinese herbal medicines of Polygonatum odoratum,Radix Isatidis,Patchouli and Echinacea was optimized,the extraction process was optimized,and Polygonatum,The effect of Radix Isatidis on the immune response of Newcastle disease and chicken avian influenza vaccine.The specific research methods and results are as follows:The Chinese herbal medicines of Polygonatum odoratum,Radix Isatidis,Patchouli and Echinacea were selected,and the extraction rate of polysaccharides was tested by water extraction,ultrasonication and enzymatic hydrolysis.The results showed that the extraction rate of Polygonatum odoratum and Radix Isatidis under ultrasonic method was higher than that of other methods.The two extraction methods were the highest(P<0.05),reaching40.35% and 6.44%,respectively,while the extracts of patchouli and echinacea were extracted using the above three extraction methods,and the extraction rates were not satisfactory.At the same time,the orthogonal test of polysaccharide extraction from Polygonatum odoratum and Radix Isatidis was carried out by ultrasonic method,and the extraction process was optimized.The results showed that the optimal extraction process of Polygonatum odoratum polysaccharide was 1:20,ultrasonic time 60 min,temperature100℃,extraction.The number of times was 3 times;the optimal extraction process of Radix Isatidis was 1:20,the ultrasonic time was 30 min,the temperature was 90℃,and the extraction times were 2 times.The chicks were used as experimental animals to carry out the experiment on the immune effect of the Newcastle disease vaccine of Polygonatum odoratum polysaccharide and Radix Isatidis as the immune enhancer.The chicks were divided into 6 groups forimmunization,which were blank test group,Polygonatum polysaccharide group,Radix isatidis group and ring.Phosphoramide group,Polygonatum odoratum polysaccharide plus cyclophosphamide group,Radix isatidis polysaccharide plus cyclophosphamide group,antibody monitoring was performed on the 0th day before immunization,7,14,21,28,35 d after immunization.The results showed that 35 days after immunization,The antibody titer of Polygonatum odoratum polysaccharides was significantly increased under immunosuppression(P<0.05),and there was no significant difference in serum Ig A antibody,peripheral lymphocyte transformation rate and organ immune index(P>0.05).The immune effect test of chicken avian influenza vaccine was carried out with Polygonatum odoratum polysaccharide and Radix Isatidis polysaccharide as adjuvant.The blank control group,vaccine control group,Polygonatum polysaccharide adjuvant group and Radix isatidis polysaccharide adjuvant group were used for immunization,0 d before immunization.Serum was collected at 7,14,21,28 and 35 d after immunization,and HI was detected on D7 antigen and r D8 antigen,respectively.The results showed that the antibody titer of D7 antigen was significantly increased on the 21 st day after immunization and 28 days after immunization in the Polygonatum polysaccharide adjuvant group and Radix isatidis polysaccharide adjuvant group(P<0.05),21 days after immunization and 35 days after immunization.The antibody titer of r D8 antigen was significantly increased(P<0.05).In summary,the best method for extracting polysaccharides from Polygonatum odoratum and Radix Isatidis is ultrasonic method,and orthogonal test is used to screen out the best extraction process.Polygonatum odoratum polysaccharide can be used as an immunopotentiator to enhance the immune enhancement effect of Newcastle disease vaccine.Polysaccharide and Radix Isatidis as vaccine adjuvants can significantly increase the antibody titer level of avian influenza vaccine. |
