Chicken coccidiosis is caused by a variety of Eimeria coccidia parasite in the intestinal epithelial cells of chickens.The incidence rate of this disease is high,with wide distribution range and serious damage,causing serious economic losses to the chicken industry.For a long time,the use of chemical drugs is the mainstream strategy for controlling chicken coccidiosis.Chemical drugs are used to control chicken coccidiosis,which is highly likely to lead to coccidian resistance as well as the residues of chicken anticoccidial drugs,thus affecting human health.Although the addition of various anticoccidial drugs to the feed can prevent and control coccidiosis,but which often causes the drug resistance of coccidian and drug residues and other problems,this has been one of the challenges facing the use of chemical drugs.Chinese herbal preparations is used as one of the effective strategies to prevent and control chicken coccidiosis,with the advantages of not being easy to produce drug resistance,small side effects and the others,and which has a good application prospect.In this paper,the in vitro passage cell screening method was used to determine the monomer of effective natural compound and select the corresponding effective prescription of Chinese medicine composition for the verification of animal experiment.First,it collected fecal(cushion)samples from chicken farms of different scales in parts of southern China(mainly in Guangdong Province),to collect coccidian strains from different geographical sources for the resistance investigation of anticoccidial drug.Due to the particularity of sampling,all the sampled chicken farms were the onset chicken farms.Due to the sampling with targeted aim,it resulted in a high positive rate.Except for two places of Jieyang in Guangdong and Yunfu in Guangdong,the positive rate of chicken coccidian in other areas was all 100%.All of them are mixed infections,among them,the infection rate of Eimeria tenella was 67%,and the infection rate of Eimeria acervulina was65%.These two types were predominant in infection and belong to dominant species.According to oocyst strength statistics,the proportion of the sample with the infection intensity of“++++”was 42%,the proportion of the sample with the infection intensity of“+++”was 21%,the proportion of the sample with the infection intensity of“++”was21%,the proportion of the sample with the infection intensity of“+”was 17%,and the oocyst strength was high.According to the analysis of drug resistance of coccidian in 11kinds of samples used more drugs currently,the degree of drug resistance to coccidian was serious,and the same strain of strains is resistant to multiple drugs at the same time.The drug resistance of drug itself was serious,and through the results,it could be seen that the drug resistance to Lasalocid sodium was relatively mild and chicken coccidian was relatively sensitive to it.At the same time,Eimeria tenella MDBK cell culture system was used to screen monomer compounds,to obtain Artemisinin,Cinnamaldehyde and Halofuginone with better effect of anticoccidial growth on the cells.Among the inspected samples,the IC50values of Artemisinin and Halofuginone were 7±0.6μM and 2±0.3μM,respectively,with good sensitivity.Due to the corresponding Chinese medicine ingredients,the corresponding Chinese medicines were purchased.And according to the characteristics of chicken coccidiosis and the contraindications of the pharmacopoeia,and according to the active ingredients of Chinese medicine and the principle of streamlining and high efficiency,three traditional Chinese medicine prescriptions were formed,through the observation to the clinical effect of treating coccidiosis,it conducted the screening test for anticoccidial prescriptions.Through Chinese herb to conduct the compound anti coccidian effect test in the animal experiment,prescriptions 1(Cinnamon 20,Rhubarb 10,Artemisia apiacea 20,Agrimony 18,Humus flava usta 18,Dried tangerine peel 18)and Prescription 3(Pulsatilla1 and Agrimony 1,Artemisia apiacea 1)were screened,with excellent effects.And when it was used with a dose of 1 g per day for each chicken and relatively better effect on the day of administration for attacking the worm was obtained,with a better effect against Eimeria tenella.The ACI values of Prescription 1 and Prescription 3 were 165.13 and 173.90,respectively,which were compared to the ACI value of 169.01 in the Sulfachloropyrazine sodium of the drug control group,the effect were the same,and they could basically replace the use of Sulfachloropyrazine.The ELISA kit was used to determine the level changes of cytokine IFN-γ,IL-2 and IL-12 in chicken serum at various stages.The traditional Chinese medicine prescription did not have much effect on the cytokine levels in serum.Seen from the clinical pathological tissue section,there was better effect of anti coccidian on the day of administration for attacking the worm.According to the obtained pathological section,the application of administered drugs before two days of attacking worm would have a slight inflammatory reaction to the intestinal tract of the chicken.The analysis of this reason might be that the prescription would cause slight damage to the intestinal tract of the chicken,which could cause the form of diarrhea to destroy the intestinal tract due to the effect of anti coccidian.
