In this experiment,Henan local goat breeds in white goat as the research object,based on the variety of slaughter performance and meat quality indexes,amino acids,fatty acids and related gene expression were determined,to discuss the Henan local goat breeds in white goat production performance,meat quality and Germplasm characteristics.It is divided into the following three tests:Tast one: 15 Yaoshan white goats by weight divided into A(33.948 + 1.6186 kg),B(37.912 + 1.8115 kg),C(45.234 + 4.3231 kg)three groups of relationship between body weight and the body size index analysis.The results show that the A group bust was significantly lower than that in B group and C group(P < 0.01);group C tare,bone weight,carcass weight,net meat weight was significantly higher than that of group A(P < 0.01);group C,slaughter rate,net meat rate,skin area was significantly higher than that of group A(P < 0.01).C grou P of skin width significantly higher than that of group A(P < 0.01),significantly higher than that of group B(P < 0.05),three groups of mesothelial length were not significant,indicating the skin area with the increase of the weight increase is mainly caused by the change of the width of the skin.These results indicate that with the increase of sheep body weight,body height,chest circumference,skin width,skin area,weight and carcass weight,slaughter rate,net meat weight,bone weight,net meat rate increased in different degrees.In this experiment,10 sheep with similar body weight were selected for meat quality analysis of three muscles,including triceps,longissimus dorsi and gluteal muscles.The results showed that p H/0h and p H/24 h of muscle were in the range of normal mutton.There was no significant difference in the rate of cooked meat,p H/0h,p H/24 h,and shearing in the triceps,the longest muscle in the back and the gluteal muscles.There was no significant difference in the diameter of muscle fibers between the three kinds of muscles,but the muscle fiber density of the gluteal muscles was significantly greater than that of the arm triceps(P < 0.05).Tast two:10Yaoshan white goats were selected,and the longissimus dorsi,triceps and gluteus were collected after slaughter,and the contents of conventional nutrients,fatty acids and amino acids were determined.The results showed that the gluteal muscles protein content was significantly higher than that of the triceps(P<0.01),and it was higher than that of the longissimus dorsi(P=0.05).The muscles fat content of the longissimus dorsi was significantly higher than that of the triceps and gluteus muscles(P<0.01).Gas chromatograph was used to detect the content of fatty acids in muscle samples,and 16 kinds of fatty acids were test in total.Among them,C18:1 has the highest content,followed by(C16:0)and(C18:0).The contents of muscle fatty acids in the three regions were significant different except C18:2,C20:3n3 and C24.Among them,C18:0 and C18:1 in the longissimus dorsi were significantly higher than in the triceps and gluteus muscles(P<0.01).C16:0 and C14:0contents was significantly higher than that in the trice Ps(P<0.01),also significantly higher than the gluteus muscle(P<0.05).This study using high Performance liquid chromatography(HPLC)tested the contents of amino acids,the proportion of different amino acid content varied in different parts of the muscles,but total amino acid contents have no significant difference between the three parts of the muscles.The ratio of EAA/NEAA in the triceps,the longissimus dorsi and the gluteus respectively were 1.171±0.1101、1.1285±0.0694 and 1.2061±0.0596,and the ratio is much higher than 60%.EAA/TAA respectively were 0.5385±0.0234、0.5298±0.0152 and 0.5465±0.0123,and more than 40%.Test three:In this experiment,Yaoshan white goat as the research object,using RT-PCR technology,analysis the relative expression of the triceps muscle of arm,longissimus muscle,muscle and so on of Myo D,IGF-1 and Myo G,MSTN,and Smad4 gene.The results showed that the relative expression of Myo D,IGF-1 and Myo G genes in the triceps brachii muscle was the highest,and they were significantly higher than those in the gluteal muscles(P < 0.05).The relative expression of MSTN and Smad4 genes in the gluteal muscles was the highest,and was significantly higher than the relative expression of MSTN and Smad4 genes in the arm triceps(P < 0.05). |
