ObjectivesTo explore self-made QiLan Compound effects on Immunosupp ressive model mice immune function by using Cyclophosphamide.The thesis clarifies preliminarily that the mechanism of action about the immune regulating,meanwhile,it provides theory basis about the using of QiLan Compound,and a reference for disease prevention a nd control of veterinary clinical immunosuppression.Methods(1)108 KM mice were randomly divided into 6 groups:blank group(NCG),model group(CYG),and different doses of QiLan compound group(QL1G、QL2G、QL3G)and positive control grou p(LMSG).NCG was Intraperitoneally injection normal saline and the remaining groups were intraperitoneally injected with cyclophosp h-amide(CY)80 mg/kg·BW for 3 days.NCG and CYG were gavaged with distilled water,QL1G-QL3G were gavaged with 4g/kg·BW,8g/kg·BW and 16g/kg·BW of QiLan compound decoction,and LMSG was gavaged 20mg/kg·BW of levamisole(LMS),for 14 days.Separately detected the following indicators:weight,feed intake,blood routine,plasma immunoglobulins(IgG,IgM,IgA),complement(C3,C4),and cytokines(IL-2,IL-4,IL-8,IL-10,IL-13,TNF-α,IFN-y)content,immune organ index,spleen lymphocyte proliferation rate,macrophage phagocytic index,peripheral blood cell phenotype(CD4+/CD3+)and antioxidant index(T-AOC,T-SOD,GSH-Px,MDA).(2)60 KM mice were randomly divided into 6 groups:blank group(NCG),model group(CYG),and different doses of cell counts,tight junction protein ZO-1 expression,intestinal slgA content,and TLR4/Mdy88/NF-κB pathway changes.Results(1)①Compared with NCG,the body final weight,weight gain,fodder,the macrophage phagocytosis index,T and Bcell proliferation,the content of cytokines,immunoglobulin and complement in CYG model group were extremely singnificantly decreased(p<0.01),and the immune organ index were extremely significantly increased(p<0.01),This shows that the immunosuppression model was successfully constructed.②Compared with CYG,the body final weight and weight gain of QL1G~QL3G were extremely singnificantly increased(p<0.01),the body final weight and weight gain of LMSG was singnificantly increased(p<0.05);the spleen index of QL1G~QL3G and LMSG were singnificantly decreased(p<0.05),but the thymus index were extremely singnificantly decreased(p<0.01).③Compared with CYG,T cell proliferation of QL2G was singnificantly increased(p<0.05),T cell proliferation of QL3G was extremely singnificantly increased(p<0.01),B cell proliferation of QL1G~QL3G and LMSG were extremely singnificantly increased(p<0.01),the clearance index of QL2G and LMSG were singnificantly increased(p<0.05),the clearance index of QL3G was extremely singnificantly increased(p<0.01),the phagocytic index of QL1G was singnificantly increased(p<0.05),the phagocytic index of QL2G、QL3G and LMSG were extremely singnificantly increased(p<0.01).④Compared with CYG,the content of immoglobulin in QL1G~QL3G and LMSG were extremely singnificantly increased(p<0.01),the content of complement in QL1G was singnificantly increased(p<0.05),the content of complement in QL2G、QL3G and LMSG were extremely singnificantly increased(p<0.01);the content of IL-2 in QL1G~QL3G and LMSG were extremely singnificantly increased(p<0.01),the content of IL-4、IL-10、IL-13 in QL1G was singnificantly increased(p<0.05),the content of IL-4、IL-10、IL-13 in QL2G、QL3G and LMSG were extremely singnificantly increased(p<0.01);he content of IFN-γin QL1G、QL2G and LMSG were singnificantly increased(p<0.05),the content of TNF-αin QL2G、QL3G and LMSG were singnificantly increased(p<0.05).⑤Compared with CYG,the content of MDA in QL1G~QL3G、LMSG were extremely singnificantly reduced(p<0.01),the active of T-AOC、SOD、GSH-Px in QL3G was extremely singnificantly increased(p<0.01).(2)①Compared with NCG,the Villi height and V/C ratio in CYG mice was extremely significantly reduced(p<0.01),the crypt depth in CYG mice was extremely significantly thickening(p<0.01);Compared with CYG,the Villi height and V/C ratio in QL1G~QL3G and LMSG have extremely significantly increased(p<0.01),the crypt depth has extremely significant thinning(p<0.01);QL12G-QL3G groups have a dose-dependent increase in villus length and V/C ratio.②Compared with NCG,the number of GC in intestine of CYG mice was extremely significantly reduced(p<0.01);Compared with CYG,the number of GC in the intestine of QL1G~QL3G and LMSG mice extremely significantly increased(p<0.01).The relative expression of ZO-1 in NCG was extremely significantly higher than that in CYG,QL1G-QL3G and LMSG(P<0.01).Compared with CYG,the relative expression of ZO-1 in QL1G-QL3G was significantly increased(P<0.05).③Compared with NCG,the expression of ZO-1 protein was lower in CYG.Compared with CYG,the protein levels of QL1G-QL3G and LMSG increased with increasing concentrations.④Compared with NCG,the secretion of TLR4/Mdy88/NF-κB signaling pathway in CYG was extremely significantly decreased(p<0.01).Compared with CYG,the secretion of TLR4 and Mdy88 signaling pathway was extremely extremely significantly increased in QL2G、QL3G mice(P<0.01);in the level of NF-κ B factor,QL1G-QL3G mice have a extremely significantly higher increase than CYG(P<0.01).⑤ Compared with NCG,the SIgA content of CYG extremely significantly decreased(P<0.01).Compared with CYG,the SIgA content of QL1G~QL3G and LMSG extremely significantly increased(P<0.01).ConcolusionsQiLan compound can play a significant role in repairing immune damage in CY immunosuppressive mice from various aspects such as cellular immunity,humoral immunity,and intestinal immunity,and has potential clinical application value in the prevention and treatment of animal immunosuppressive diseases.
