Abstract/Summary:
The Chinese honeybee(Apis cerana)plays vital roles in pollination for agro-ecosystem and honey bee products in our country.The Qinling-Daba Mountain Areas which is the biggest region that Apis cerana has distribution have superior homogeneous environment.This study aims to investigate the population differentiation and among 17 Apis.cerana localities across the Qinling-Daba Mountain Areas combined with two extra-populations populations,correlation analysis between genetic distance with geographical distance,and to explore the possible factors contributing to genetic differentiation and genetic diversity based on 22 microsatellite loci.(1)The genetic differentiation(Fst)and genetic distances(Da)among 17 Apis.cerana localities across the Qinling-Daba Mountain Areas were 0.0052~0.033,0.034~0.146,respectively.Up to 95%of the variation was observed within sampling localities while less variation between localities.Combine with a variety of analytical methods indicating that no population differentiation was detected;(2)Apis cerana of Qinling-Daba Mountain Areas had no population genetic differentiation.There are 650 km between the two localities that has farthest distance.Correlation analysis between genetic distance with geographical distance achivied equation of linear is y=-0.00131x+0.02474,R~2=0.02328s indicating that 650 km con’t cause population differentiation and there is no relativity between genetic distance and geographical distance in this environment such as Qinling-Daba Mountain Areas.(3)There were three kind of genetic polymorphism in the 22 microsatellite loci that 16 is highly polymorphiclocus loci(PIC>0.5),3 is moderate polymorphiclocus loci,and 3 is low polymorphiclocus loci.There are abundant gengetic diversity in Qinling-Daba Mountain Areas.The mean expected heterozygosity(He),observed heterozygosity(Ho),number of alleles(Na),number of effective alleles(Ne),polymorphism information content(PIC)and Shannon’s information index(Ⅰ)were 0.599±0.015,0.555±0.019,7.329±0.513,3.460±0.155,0.560±0.015 and 1.287±0.041 respectively.
