Abstract/Summary:
Liver cancer is one of the cancers with high morbidity and mortality rates worldwide.Early-stage liver cancer can be treated by surgery.However,because the incidence rate of liver cancer is not easy,the common cancer is usually advanced.For the unsuccessful surgical treatment of liver cancer,targeted therapies and immunological drugs have been developed and combined with chemotherapy.However,these treatments are usually difficult to bear with the high cost of treatment.Therefore,we want to find potential small molecule metabolites that can judge the sensitivity of patients to immunotherapy by analyzing the blood samples of patients with liver cancer before treatment and applying them in clinical diagnosis in the future to save treatment costs for patients.Diabetic retinopathy is one of the major complications of diabetes.Diabetic retinopathy will cause vision loss.Photobiomodulation therapy is a new treatment method in recent years,which inhibits retinopathy in a non-invasive way.This treatment method is cheap and easy to operate.The focus is that no adverse reactions of treatment have been observed,but the regulatory mechanism of photobiomodulation therapy is not fully understood.The changed small molecule metabolites are found through the analysis of mouse serum by nontargeted metabolomics.This may be the metabolic mechanism affected by photobiological regulation,which provides a research basis for subsequent proteomics or genomics.The main research contents of this paper are as follows:1.Apply liquid chromatography and mass spectrometry to analyze the metabolites in the serum of patients with liver cancer.Judge the tumor progression after immunotherapy according to the evaluation standard of solid tumor efficacy,and divide the patients into three groups: partial reaction,stable disease,and progressive disease.Through the detection of small molecule metabolites in the serum of patients with liver cancer,combined with partial least squares discriminant analysis,the serum metabolites of the three groups of patients were analyzed,and the metabolites with great differences were analyzed and identified.Through a group of biomarkers identified,a model for predicting the efficacy of liver cancer immunotherapy was established.This method has potential clinical application value for patients to choose immunotherapy.2.Strategy of nontargeted metabolomics analysis of serum metabolites based on liquid chromatography was established by establishing a mouse model(normal group,diabetic model group,and photobiological regulation diabetes model group).The mass spectrometry acquisition mode is positive ion mode.Combined with principal component analysis and partial least squares discriminant analysis,two sets of data analysis methods are used to find a large set of lipid biomarkers.These biomarkers have different signal intensity in three groups of models.Generally speaking,the signal value in the normal group is high in the diabetic model group,but the signal in the photo biologic regulation group is higher than that in the diabetes group.Lower than normal group signal.Conversely,if the signal value is low in the normal group,the signal in the diabetic model group is high,but the signal in the photo biologic regulation group is lower than that in the diabetes model group and higher than that in the normal group.This method proves that photobiological regulation affects lipid metabolites in mice,which may have beneficial effects on the retina.
