In recent years,the research on carboxylic acid-based coordination polymers has been in-depth development.As one of the most important groups in the synthesis of coordination compounds,carboxylic acid groups are The unique properties of carboxylic acid complexes.They make the complexes have a high coordination number,which makes complex and novel complex structure changes,and plays a very important role in coordination chemistry.In addition,lanthanide metals have low toxicity to the human body,which provides the feasibility for its biochemical research,and has also attracted the focus of many anti-cancer drug researchers.In this article,3,5-bis(3-carboxyphenoxy)benzoic acid is used as the ligand(hereinafter referred to as L)and N,N-dimethylformamide(DMF)sed as the solvent to synthesize the complex by solvothermal method.1-3,the molecular formula is[La(L)2(DMF)2]n(1),[Ce(L)2(DMF)2]n(2),[Er(L)2(DMF)2]n(3).During its characterization and cytotoxicity research,the following work was carried out:1.Use X-ray single crystal diffractometer to determine the crystal structure of complex 1-3,belonging to the asymmetric triclinic system,located in the P-1 space group,irregular prism geometry,with a two-dimensional planar structure;2.The elemental analysis of complex 1-3 was carried out to determine the elemental composition of complex 1-3;PXRD and infrared spectroscopy were used to characterize its structure,and the phase purity of the synthesized product was analyzed;3.Use ultraviolet and fluorescence spectroscopy to detect the ability of complexes1-3 to bind to DNA,use calf thymus DNA(CT-DNA)to analyze the interaction mode of complexes with CT-DNA,and use Stern-Volmer equation to calculate fluorescence quenching Constant KSV;4.Test by agarose gel electrophoresis experiment and determine the ability of complex 1-3 to cut p BR322 plasmid DNA,and analyze after obtaining the experimental data;5.The ability of complex 1-3 to induce apoptosis of He La cells was detected by flow cytometry,and several experiments were set up to verify its ability.The killing effect of complex 1-3 on He La cells was tested by fluorescence inverted microscope,and its mechanism was analyzed.The results show that the complexes 1-3 with new structure synthesized in this paper are metal-organic complexes with two-dimensional planar structures,and all of them can interact with DNA,thus affecting the stability of DNA.They have good apoptosis-inducing effect on He La cells and good cytotoxicity,and can be used as promising anticancer drug materials.
