Sea buckthorn fruit oil resources is a kind of medicine and food plants.Sea buckthorn fruit oil has the main ingredient palmitoleic acid,which has the functions of enhancing immunity,anti-tumor,protecting liver,anti-inflammatory,and alleviating various metabolic diseases.in order to better develop its effective components,increase its utilization value,and further improve the efficiency of the comprehensive utilization of sea buckthorn fruit oil and the added value of products,this paper studied the extraction of palmitoleic acid from sea buckthorn fruit hair oil by oil refining,urea embedding and molecular distillation,and the effects of sea buckthorn fruit oil and palmitoleic acid on hyperlipidemia rats.The main results are as follows:(1)Sea buckthorn fruit crude oil was used as raw material for oil refining,and the process of degumming and decolorization of sea buckthorn fruit crude oil was optimized by ultrasonic coenzyme method.The degumming process was established by using the amount of enzyme,ultrasonic time,ultrasonic temperature and ultrasonic power as the influencing factors,and the phospholipid content as the response value.The results showed that the amount of enzyme was 5%,the ultrasonic time was 39 min,the ultrasonic temperature was 50℃,and the ultrasonic power was 180 W,and the phospholipid content of sea chinkaw fruit oil was 0.097 mg/g.The decolorization process was established by using decolorization temperature,decolorization agent amount and decolorization time as the influencing factors,and the decolorization rate as the response value.The results showed that the decolorization temperature was 145℃,the decolorization agent amount was 3%,and the decolorization time was 26 min.The decolorization rate of the obtained decolorization fruit oil was 92.11%.After oil refining,the oxidation stability of sea buckthorn fruit oil was reduced.The compound antioxidant was 0.02% BHT+0.01% citric acid.The refined sea buckthorn fruit oil has better fluidity,lower phospholipid content,lower impurities in the degummed oil and lighter color,which is very beneficial to the storage of sea buckthorn fruit oil and the application of downstream users.(2)After refined sea buckthorn fruit oil,palmitoleic acid is enriched by urea embedding method,refined sea buckthorn fruit oil is embedded by urea embedding method,the embedding time,the ratio of urea to substrate,and the ratio of solvent to urea are screened out as influencing factors.The palmitoleic acid content is the response value,and the Box-Behnken method is used to optimize the design to establish the regression equation model and perform response surface analysis.The results showed that the embedding time was 13 hours,the ratio of urea to substrate was 2.5∶1,and the ratio of solvent to urea was4∶1.The yield of the product was 47.52%,and the content of palmitoleic acid was 56.40%.Provide a theoretical basis for the extraction process and content of palmitoleic acid.(3)On the basis of urea embedding and enriching palmitoleic acid in sea buckthorn fruit oil,short-range molecular distillation was further used to enrich high-concentration palmitoleic acid.The distillation temperature,rotor speed,and feed rate were screened out as influencing factors.The palmitoleic acid content was For the response value,the Box-Behnken method is used to optimize the design to establish a regression equation model and perform response surface analysis.The results showed that the distillation temperature was 90℃,the rotor speed was 110 r/min,the feed rate was 1 m L/min,and the obtained palmitoleic acid content was 67.56%.It can provide a theoretical basis for the further extraction research of high-concentration palmitoleic acid,and at the same time expand the source of palmitoleic acid,which is conducive to the indepth development of sea buckthorn and improves its utilization rate.(4)The hyperlipidemia model rats were given sea buckthorn fruit oil and sea buckthorn fruit oil combined with palmitoleic acid by intragastric administration.The results showed that compared with model group,the serum concentrations of TC,TG and LDL in each dose group were significantly decreased(P<0.05),while the concentration of HDL was significantly increased(P<0.05).Significantly increased the antioxidant index GSH content and SOD(P<0.05)activity in serum,and significantly decreased the content of MDA(P<0.05);Pathological section results showed that sea buckthorn fruit oil combined with palmitoleic acid group significantly reduced steatosis,liver fat vacuolation,liver cell injury and liver lipid accumulation.In conclusion,sea buckthorn fruit oil combined with palmitoleic acid has a strong protective effect on hyperlipidemia rats.The results showed that the content of phospholipid in refined sea thorn fruit oil was 0.097 mg/g,the decolorization rate was 92.11%,and the compound antioxidant was 0.02% BHT+0.01% citric acid.The content of palmitoleic acid obtained by urea embedding was 56.40%.The content of palmitoleic acid was67.56% by molecular distillation.Sea buckthorn fruit oil combined with palmitoleic acid had a strong protective effect on hyperlipidemia rats.The content of palmitoleic acid was increased by oil refining,urea embedding and molecular distillation,and related animal experiments were conducted to provide reference for the development of effective components of sea buckthorn fruit oil,the improvement of comprehensive utilization value,and the industrialization of palmitoleic acid.
