| Melanin is a biological pigment produced by tyrosine under the catalysis of tyrosinase with a series of biochemical reactions,and stored in melanocytes in the form of aggregation.In nature,most plants and animals take place in this biochemical reaction and produce this biopigment,which is responsible for the various colors of living things.The senile spot and freckle formed by excessive accumulation of melanin is not only an important sign of airframe aging and skin aging,but also an important factor affecting airframe health.The ubiquitous natural antioxidants and some synthetic antioxidants have antioxidant functions,which can improve immunity and maintain health of organisms by effectively removing reactive oxygen species in the body.This study by building in vitro melanin formation model to explore the different wavelength of light source,as well as natural and artificial antioxidants to remove active oxygen free radicals of melanin formation in vitro and in vivo.then the zebrafish as a model organism which is to further explore the natural antioxidant and synthetic antioxidants restrain melanin formation mechanism.This paper provides theoretical basis for the study of whitening and anti-aging products and health care products,and has important guiding significance for promoting the development of cosmetics and health care products industry.The main work and results are as follows:(1)In vitro melanin model construction: The best simulation conditions were determined based on the reaction of Mushroom tyrosinase catalyzed L-tyrosine(Tyr).On this basis,the effect of different wavelengths of light sources on the formation of melanin was explored,and the scavenging effect of natural antioxidants(AA,GA,GSH)and synthetic antioxidants(BHA,BHT,PG)on the scavenging effect of reactive oxygen radicals on melanin in vitro was explored.The results shows that the formation rate of melanin is the fastest under UV light,while the formation rate of melanin is the slowest under blue light.That is the shorter the wavelength,the faster the formation rete of melanin.Among the natural antioxidants,glutathione(GSH)has the most obvious inhibition effect on melanin formation system,while among the synthetic antioxidants,propyl gallate(PG)has the most obvious inhibition effect on melanin formation system.The results shows that GSH(natural antioxidant)and PG(synthetic antioxidant)have the most significant inhibitory effects on melanin formation.Therefore,GSH and PG are selected to further explore the effect of antioxidants on melanin formation in vivo.(2)In vivo inhibition of melanin by GSH and PG.In this study,Zebrafish were used as model biological model,and the zebrafish were fed with GSH and PG.After a period of time,the content of melanin in zebrafish was measured by enzyme plate analyzer to evaluate the inhibitory effect of GSH and PG on melanin in zebrafish.The results shows that GSH and PG at ≤2.5 mmol/L and 0.25 mmol/L don’t affect the survival rate,hatchability and malformation rate of zebrafish embryos,and inhibited melanin formation in zebrafish to some extent.On this basis,the reversible effects of GSH and PG on inhibiting melanin formation in zebrafish are further explored. |
