The Pleurotus djamor as a cherished mushroom,has excellent nutritional value and its main component polysaccharide has been proven to have various physiological activities,such as antioxidant,anti-aging and anti-cancer.However,most of the P.djamor products available in the market are primary processed products,so it is extremely urgent to develop a further processed product.In this study,we determined the appropriate amount of zinc acetate to be added to the liquid medium and the optimal extraction parameters of P.djamor zinc polysaccharide(PZP),and the formulation of P.djamor zinc polysaccharide chewable tablets(Zn-PCT)were optimized,and the in vitro antioxidant activity of Zn-PCT was examined to verify its repairing effect on reproductive damage induced by nonylphenol(NP)in mice.The main results were as follows:(1)Under liquid culture conditions,the appropriate amount of zinc acetate to be added to the medium was 50 mg/L.The optimal extraction conditions for PZP were 30.99 water addition,2.46 h extraction time and 15.29 h alcoholic sedimentation time,and the theoretical yield of zinc polysaccharide from P.djamor was 1.06 %.According to the actual operability,the factors were adjusted to 31.00 water addition,2.5 h extraction time and 15 h alcoholic sedimentation time,and the theoretical predicted yield was 1.05 %,and the actual value was 1.04±0.03 %.(2)The best formula was PZP 25.0 %,xylitol 25.0 %,citric acid 1.5 %,magnesium stearate1.0 %,mannitol 47.5 %,the resulting chewable tablets had smooth surface,delicate taste,sweet and sour,and moderate hardness.(3)Zn-PCT showed excellent in vitro antioxidant capacity by scavenging hydroxyl radical,superoxide anion and DPPH radical at a concentration of 1.0 mg/m L,reaching 39.1±1.07 %,22.6±1.69 % and 1.8±1.89 %,respectively.(4)Efficacy of Zn-PCT on NP-induced reproductive injury.It can significantly improve the decrease of testicular coefficient produced by NP-induced in mice and reduce the tissue structure damage of testis and epididymis.At the sperm level,Zn-PCT alleviated the reduced sperm density and increased malformation rate brought about by NP.For sex hormone levels in mice,testosterone levels in mice serum rebounded after Zn-PCT treatment.Zn-PCT enhanced the activities of glutathione peroxidase(GSH-Px),catalase(CAT)and superoxide dismutase(SOD),and reduced the levels of malondiadehyde(MDA)and reactive oxygen species(ROS),and alleviated the damage caused by oxidative stress.The level of apoptosis in the testis was alleviated after treatment with Zn-PCT,the level of B-cell lymphoma-2 protein(Bcl-2)was elevated by 91 %,and the expression level of Bcl-2 associated x protein(Bax)was decreased by about 21 %.Zn-PCT effectively reduced the expression levels of cyclooxygenase-2(COX-2),gamma interferon-?(IFN-?),tumor necrosis factor-?(TNF-?),interleukin-6(IL-6),and other proteins in the testes of NP-exposed mice by about 55 %,51 %,49 %,and 76 %,respectively,compared with the model group.In conclusion,Zn-PCT can alleviate reproductive damage in male mice by reducing testicular tissue damage,improving the antioxidant capacity of the body,increasing androgen levels,inhibiting testicular cell apoptosis,and reducing the expression of inflammatory factors in the testis.
