Betacyanin is one of the natural food pigment,and it is widely present in many plants such as Polygonaceae,Chenopodiaceae,Mirabilis,Pokeweed and other plants.It belongs to ketone and quinone derivatives,and has blood lipid-lowering and anti-cancer properties.It is easy to be absorbed by the human body after eating,and has no side effects.However,there are few reports about the processing and utilization of Betacyanin.At present,alcoholic liver disease has become one of the common diseases that endanger human health,and the hepatoprotective effect of natural ingredients has attracted increasing attention from researchers.In the early stage of this research group,DEAE-52 cellulose was used to extract and isolate the Betacyanin concentrated juice from the red beet extract solution,and then the concentrated juice was prepared into a Betacyanin spray-dried powder(BSP).This thesis used BSP as raw material to study its protective effect on alcoholic liver injury,and prepares BSP chewable tablets,which provides a basis for the development and utilization of red beets as functional health food.The following are main results.(1)The antioxidant activity of BSP:The antioxidant activity of BSP was evaluated by DPPH,ABTS,hydroxyl free radical scavenging capacity test and total reducing power test.The result of antioxidant test showed that the antioxidant activity of BSP was dose-dependent in the range of 40-1280 ?g/mL.When the concentration was at 1280 ?g/mL,the clearance rate of BSP on DPPH,ABTS,and hydroxyl radicals was 91.19%,83.23%and 91.26%,respectively.The IC50 value of BSP scavenging DPPH free radical,ABTS free radical and Hydroxyl free radical was 109.65 ?g/mL,260.49 ?g/mL,89.59 ?g/mL,respectively,and the reducing power of BSP and vitamin C was equivalent in the range of 40 to 1280 ?g/mL.(2)The protective effect of BSP on acute alcoholic liver injury in mice was studied:The model of alcoholic liver injury in mice was established,and the protective effect of BSP on alcoholic liver injury was studied.The results showed that the liver index of dose groups was significantly reduced with BSP(0.25,0.5,1.0 g/kg)(P<0.01),plasma alanine aminotransferase(ALT),aspartate aminotransferase(AST)and triglyceride(TG)in alcoholic liver injury mice were significantly reduced with BSP(0.25,0.5,1.0 g/kg)(P<0.01).The content of glutathione(GSH)and the activity of superoxide dismutase(SOD)in liver tissues were significantly increased(P<0.01),and the content of malondialdehyde(MDA)was significantly reduced in alcoholic liver injury mice with BSP(0.25,0.5,1.0 g/?g)(P<0.01).Pathological sections showed that BSP can reduced inflammation and lipid droplets,and repair liver cells.Indicating that BSP had positive protective effect on acute alcoholic liver disease in mice.(3)Preparation of BSP chewable tablets:Wet granulation and tableting,using BSP as the raw material,determine the best formula of BSP chewable tablets through single-factor experiments.The results showed that the optimum technological formula was as follows:The added amount of raw materials(BSP)was 33.0%,the added amounts of each component in the auxiliary materials were xylitol 20.0%,mannitol 20.0%,sorbitol 15.0%,microcrystalline cellulose 6.0%and maltodextrin 6.0%,citric acid was 1.0%and magnesium stearate was 1.5%at the conditions of 75%ethanol as the wetting agent.The sensory score of the BSP chewable tablets were uniform in color,smooth and round in appearance,sweet and sour,no powder and grain,moderate hardness,and good taste.
