| BackgroundNitrate/nitrite in the diet had an impact on human health.A high concentration of nitrite had konwn about various health problems such as methemoglobinemia,respiratory diseases,neurological dysfunction and cancer.Therefore,how to prevent the harm of nitrite is worthy of attention in the environment that the country concerns food safety.ObjectiveThis study explores that the protective effect of wheat embryo globulin(WEG)againsts sodium nitrite induce poisoning in rats,which provides theoretical basis to treatment nitrite poisoning.MethodFirst,36 rats were divided control group,sodium nitrite-treated group,sodium nitrite+WEG(40mg/kg)group,sodium nitrite+WEG(80mg/kg)group,sodium nitrite+WEG(160mg/kg)group,sodium nitrite+Vitamin C(80mg/kg)group.Rats was given single does of WEG for 7 days followed by a single does of Na NO2.The changes of physiological and biochemical indexe were observed about blood,kidney and liver,which proved the protective effect of WEG in rats with acute nitrite.In addition to,this study was explored inhiniting nitrification activity(37℃,p H=3.0)of heat globulin product in hydrogenation(WGPIH),which provided theoretical basis for further development of WGPIH.Results(1)WEG protect against sodium nitrite-inducein poisoning in rat bloodWEG could significantly improve the mental state and activity of rats after nitrite poisoning.Compared with the model group,dose of WEG group and vitamin C group in methemoglobin(Met Hb)levels was significantly decreased(P<0.05).About detection of serum,the levels of nitric oxide(NO),nitrite,alanine aminotransferase(ALT),aspartate transaminase(AST),blood urea nitrogen(BUN),lactate dehydrogenase(LDH)and alkaline phosphatase(ALP)in the model group were more than control group.The contents of(alkaline phosphatase)AKP,(acid phosphatase)ACP,(crcreat-inine)Cre,H2O2and(malondialdehyde)MDA were increased(P<0.05).The content of serum in WEG groups and vitamin C groups were decreased(P<0.05).Compared with the control group,the activity of-SH,(superoxide dismutase)SOD,(catalase)CAT and(gluthione)GSH in the model group were decreased(P<0.05).Moreover,the content of WEG group and vitamin C group were increased(P<0.05).(2)WEG protect against sodium nitrite-inducein poisoning in rat kidneyCompared with the control group,the kidney index of nitrite poisoning group was significantly increased(P<0.05).The contents of SOD,GSH and total antioxidant capacity(T-AOC)in kidney were increased(P<0.05).The production of MDA was decreased(P<0.05).The renal index was significantly decreased(P<0.05).The contents of Caspase-3 and Caspase-8 in kidney were decreased(P<0.05).The degree of renal degeneration and necrosis was significantly reduced.(3)WEG protect against sodium nitrite-inducein poisoning in rat liverCompared with control group,the kidney index of nitrite poisoning group was significantly increased(P<0.05).The contents of SOD and GSH in liver were increased(P<0.05).The contents of MDA of liver were decreased in WEG groups and vitaminnn C groups(P<0.05).The pathological changes of liver degeneration and necrosis were significantly reduced.(4)Nitrite Scavenging and Nitrosamine Synthesis Blocking of WGPIHThe optimum concentration of WGPIH was 200μg/m L and 120 min.WGPIH was less effective than vitamin C in inhibiting nitrosation.ConclusionsWEG could alleviate the physiological state of rats and improve endocrine disorders,the ability of anti-oxidative stress and the antiinflammatory factors and proinflammatory factors after nitrite poisoning,which had protective effect in rats with acute nitrite poisoning.It was found that WEGPIH could scavenge nitrite and block the synthesis of nitrosamine in simulated gastric juice(37℃,p H=3.0)in vitro. |
