Study On Nutritional Characteristics And Antioxidation, Inhibition Nitrosation Activities Of Wheat Seedlings

The wheat seedings are young leaves of Triticum aestivum L.,which are rich in protein,vitamins,minerals,active enzymes and so on.The wheat seedings are not only rich in nutritious substance,but also have the medicinal health effect.The wheat seedling juice is also revered as alkaline plant resources of the most close to the human body which is Nutrient-rich,balanced,safe.Predecessors' researches on wheat seedling nutrient are mainly directed at one or several related projects.In this study,the main nutrient and active ingredient contents of 4 cultivars of wheat seeding(W18,W251,W352 and W596)were detected by Chemical analysis and Spectral analysis method.And then,we explored antioxidant capacity by scavenging free radical and reducing power test,and inhibiting nitrification activity by clearing nitrite and blocking the synthesis of ammonium nitrite activity.At last,SOD of wheat seedings has been purificated,and enzymatic properties were preliminarily investigated.The main test results are as follows:1?The main nutrient contents of wheat seedlings.The constant nutrient contents of wheat seedlings are as follows: 89.819 ~ 93.326% water,1.806 ~ 2.628% reducing sugar,30.847 ~ 42.560 mg / 100 g vitamin C,4.495 ~ 5.495% soluble solids and 1.816 ~ 2.032%ash.The main active ingredient contents of wheat seedlings are as follows: 0.758 ~ 0.876%total phenol,0.170 ~ 0.302% betaine,234.814 ~ 383.657 mg / 100 g chlorophyll,440.327 ~528.463 U / g SOD enzyme,4.495 ~ 5.495% Soluble protein.The wheat seedlings are rich in a variety of mineral elements,of which three elements zinc,iron,calcium contents are higher,are 28.9 ~ 56.0mg / kg,36.355 ~ 145.617 mg / kg and 36.355 ~ 145.617 mg / kg respectively.Wheat seedlings have rich amino acids,and the proportion is close to the ideal pattern of the human body.It is a good source of essential amino acids.The nutritional value of W352 and W596 was higher than that of W18 and W251.2?Antioxidation,inhibition nitrosation activities of wheat seedlings.The wheat seedlings have food ability in antioxidation,the higher concentration of wheat seedlings juice was,the better ability in antioxidation became.Four kinds of wheat seedlings have similar antioxidant.They were good at scavenging cation and bad for scavenging anions,among them,the scavenging superoxide anion and DPPH· free radicals ability of W352's was strongest,the scavenging activities on ABTS+· and reducing power ability of W596 was the strongest.Wheat seedlings have ability to inhibit nitrification,the best effect for the W596 wheat seedlings,followed by W352,W18,W251 slightly worse,the concentration of W596 was 6mg / mL,nitrite clearance rate as high as 66.45%,when the concentration of 12 mg / mL,the blocking rate of nitrite synthesis reached 37.14%.The results show that the active ingredient content is positively correlated with its antioxidant and nitrification inhibition.3?The purification and enzymatic properties of SOD from Wheat seedings.In this research,SOD,an antioxidant component from wheat seedlings was extracted by the phosphate buffer,and the optima extraction proce of superoxide dismutase are the ratio of m/v 1?5;extraction time 2h;the heat treatment temperature 55?;the heat treatment time45 min.SOD was purifid by sulfate fractional precipitation,DEAE-52 Ion Exchange Chromatography and Sephadex G-100 gel filtration chromatography,Coomassie brilliant blue G-250 method to measure protein content.And under these conditions,the specific activity of the enzyme is 629.7U/mg.Its molecular weight is nearly 32.1 kDa.And the activity of SOD was stable at pH 5.0 ~ 8.0 and 20 ~ 70 ?,which indicated that the SOD enzyme tolerated neutral and weak acid and weak alkali environment,and had good heat resistance.It was very sensitive to KCN,H2O2,no-sensitive to the chloroform-ethanol.Therefore,this SOD which had been purifid is Cu/Zn-SOD.
