| Diabetic wound is a complication of diabete.It has complicated wound microenvironment and pathological biological defects,which seriously hinder the normal healing of wounds.Therefore,it is significance to develop a bioactive wound dressing that can quickly promote the healing of diabetic wounds.Bioactive glass with high biological activity had curative effect on the diabetic wound.Based on this,this article combined sol-gel bioactive glass with antibacterial Gelatin and PCL to prepare a new nanofiber skin wound dressing and study the effects of material composition and structure on skin healing related cells.The main research contents and conclusions are as follows:First,in this paper,dimethyloctadecyl [3-(trimethoxysilyl)propyl] ammonium chloride(QAS),Gelatin,and PCL were used as raw materials to prepare Gelatin-PCL-QAS nanofiber membrane.Experiments show that the composite fiber membranes have good cell compatibility and antibacterial properties gradually increase with the increase of QAS content.Secondly,SGBG-Gelatin-PCL-QAS electrospun nanofiber membranes with different solgel bioactive glass(SGBG)ratios were prepared.Experiments show that SGBG is successfully doped into the nanofiber membrane,which improves the tensile strength of the nanofiber membrane.SGBG dissolved ions which can be quickly released from the fiber membrane and significantly promote the proliferation and migration of mouse fibroblasts.In addition,adding SGBG can increase the original antibacterial rate of nanofiber membranes.Finally,aiming at the problem that the differentiation of fibroblasts from diabetic wound to myofibroblasts is inhibited,a radially arranged nanofiber membrane containing SGBG was prepared,and the influence of SGBG and fiber morphology on cell differentiation behavior was explored.Experiments show that the radially arranged nanofiber membranes containing SGBG can mediate the directional adhesion and spread of mouse fibroblasts and up-regulate the expression of TGF-β1,promote the differentiation of fibroblasts into myofibroblasts,and promote the expression of type I collagen,It helps diabetic wounds to shrink and heal in time. |
