Shibi tea,a kind of tea made from dry leaves of Adinandra nitida,is a special wild tea in Guangxi,Guangdong,and Guizhou.Shibi tea not only has sweet taste,but also contains flavonoids and other active ingredients.At present,Shibi tea has been found to have antioxidant,antibacterial,anti-tumor,and other health effects.This study carried out the chemical composition analysis and sensory evaluation,studied the effect and mechanisms of Shibi tea and its main flavonoid component camellianin A on acute gastric mucosal injury and acute liver injury through animal models,in order to provide theoretical basis for the development and application of Shibi tea.The content of soluble sugar,tea polyphenols and free amino acids in Shibi tea was 18.84%,24.07%and 1.21%,respectively.The results of HPLC showed that 70%methanol extract of Shibi tea contained about 31.38%of camellianin A and 1.59%of camellianin B;in catechins,Shibi tea contained about 11.04%of epicatechin,while other catechin content was less than 1%,and no GC and ECG were detected.In addition,the content of caffeine in Shibi tea was 0.15%.The results of sensory evaluation showed that the color of Shibi tea was yellow green,slightly uniform;the color of tea soup was apricot yellow and bright;the aroma was fresh with bean flavor;the taste was pure,smooth,refreshing,and long-lasting;the bottom of tea was dark green,bright,even,and resistant to brewing.The results showed that Shibi tea and its main flavonoid component,camellianin A,had good gastric protective effect in mice with acute gastric mucosal injury induced by hydrochloric acid and alcohol.The mechanism of action was related to its anti-oxidant and anti-inflammatory effect.Water extract of Shibi tea(200 mg/kg and 700 mg/kg)and camellianin A(30 mg/kg and100 mg/kg)could inhibit the activation of NFκB inflammatory pathway by reducing reactive oxygen species(ROS)level in gastric tissue,and then inhibit the downstream levels of i NOS,COX-2 and pro-inflammatory cytokines TNF-αand IL-6 to reduce inflammatory injury.The results showed that Shibi tea and its main flavonoid component,camellianin A,had good hepatoprotective effect in mice with acute liver injury induced by CCl4.The mechanism of action was related to its anti-oxidant,anti-inflammatory and anti-apoptotic effect.Water extract of Shibi tea(200 mg/kg and 700 mg/kg)and camellianin A(30 mg/kg and 100 mg/kg)could significantly reduce the levels of AST and ALT in serum of model mice,protect the liver structure and function damage induced by CCl4,increase the activities of SOD,CAT and GSH-Px in liver tissue,increase the level of GSH,inhibit the increase of MDA and ROS oxidative stress induced by CCl4,and significantly activate antioxidant pathway Nrf2 and HO-1,inhibit the phosphorylation and activation of NF-κB in inflammatory pathway,inhibit the expression of pro-inflammatory cytokines TNF-α,IL-6 and IL-β;inhibit the expression of proapoptotic factor Bax and antiapoptotic factor Bcl-2.In conclusion,the results showed that Shibi tea and Camellianin A could improve the acute gastric mucosa injury and liver injury through the mechanism of antioxidant,anti-inflammatory,and anti-apoptosis,which provides theoretical basis for the health function of Shibi tea on stomach and liver.
