Wine is inevitable in people's daily life and communication,but drinking too much can do harm to the human body,it can cause chronic gastritis,peptic ulcer and alcoholic cirrhosis directly or indirectly,it can also cause severe damage to the central nervous system,causing dizziness,nausea and vomiting.Studies have shown that the pentacyclic triterpenoid saponins had a certain antioxidant activity,inhibited alcohol absorption and protected gastrointestinal liver.In this study,an acute alcoholism mouse model was established firstly,determined the time of drunkenness,sleep time,and monitored the changes of blood alcohol concentration in mice,explored for the effect of tea saponin on acute alcoholism mice.Then,the protective effect of tea saponin on the stomach and liver of alcohol injured mice were investigated according to the pathological morphology of gastric mucosa in mice,The protective mechanisms including ethanol metabolism enzyme activity,oxidative injury,gastric emptying rate and gastric mucosal factor were also discussed.Primary research conclusions are reached as follows:(1)It is found that tea saponin could reduce the alcohol content of gastrointestinal fluid in vitro simulated gastrointestinal experiments.The acute animal experiment showed that tea saponin could significantly prolong the ebriety time,shorten the sleep time,the sleep time of the mice in the high-dose group of tea saponin was 153.9 min less than that in the model group,and can effectively inhibit alcohol absorption,reduce blood alcohol concentration,at 90 min,the BAC in the medium and high dose group of tea saponin decreased by 33.1% and 35.9% respectively,at the same time,it can delay the peak of blood ethanol concentration.Therefore,the antialcoholic effect of tea saponin was verified,suggesting that tea saponin can alleviate the drunken state,and has a certain alleviate alcoholism effect.(2)The pathomorphological changes of gastric mucosa in mice induced by alcohol poisoning showed that tea saponin could significantly reduce the degree of gastric mucosal injury and protect gastric mucosa.The protection mechanism mainly includes the following aspects: 1)Tea saponin could significantly increase the gastric emptying rate,prolong the gastric emptying time,thus allowing more first-pass metabolism,enhance ethanol metabolic activity in the stomach,inhibit alcohol absorption at the same time and reduce blood alcohol concentration.2)Tea saponin could significantly increase the activity of ADH in gastric tissue,the residual rate of methyl orange in the high dose group was 49.9% higher than that in the model control group,increase the first-pass metabolism of ethanol in the stomach,and decrease the blood alcohol concentration.3)It can significantly increase the gastric mucosa NO content and improve the microcirculation of gastric mucosa in mice with alcoholism,At the same time,the content of gastric mucosal attack factor IL-8 was significantly decreased,and the inflammatory damage of gastric mucosa was reduced.And it also can reduce inflammation factor positive expression of CD68 and MPO,the gray value of TNF-? and IL-1? were significantly decreased,and the protein expression was decreased.The results suggest that tea saponin can reduce the inflammation of gastric mucosa induced by alcohol.(3)The metabolism of alcohol is mainly in liver tissues,tea saponin can effectively inhibit ALT and AST activity,improve the pathological morphological changes of liver in mice and protect the liver.The protection mechanism mainly includes the following aspects: 1)Alcohol dehydrogenase is the most important and key enzyme in alcohol metabolism,the content of ADH and ALDH increased by 86.13% and 74.52%,respectively,compared with the model group.The results showed that tea saponin can significantly improve the activity of ADH and ALDH in liver and enhance the level of alcohol metabolism,and its effect is obviously stronger than the positive control group.2)Tea saponin can enhance antioxidant levels in mice,reduce the damage of oxygen free radicals,increase SOD activity and reduce MDA content in liver,reducing oxidative damage.
