| With the improvement of people’s living standards,obesity has become an important problem endangering human health,causing a variety of diseases.Finding safe and effective ingredients from natural plants to prevent obesity has become an important aspect of research.Polyphenols exist widely in natural plants,and have been found to have a variety of biological activities.Because of their good efficacy and safety,polyphenols have become a research hotspot in the field of obesity prevention and control at home and abroad.As an important part of natural products,polyphenols have been paid more and more attention.In this paper,(Prunus cerasifera Ehrh)as materials,on the basis of studying the preventive effect of Prunus cerasifera Ehrh fruit water extract on obesity mice induced by high-fat diet,the total polyphenols of Prunus cerasifera Ehrh fruit were extracted by organic solvent method,and the main components of polyphenols and antioxidant activity in vitro were detected;the intervention effect of Prunus cerasifera Ehrh polyphenol extract on obesity mice induced by high-fat diet was further studied by in vivo model,and the preliminary evaluation was made The lipid-lowering activity of phenolic extract from Prunus cerasifera Ehrh fruit was evaluated.The research contents and main results are as follows Study on the lipid-lowering effect of Prunus cerasifera Ehrh)fruit extract in diet on obese mice1.The mice were fed with high-fat diet,and were fed with Prunus cerasifera Ehrh fruit water extract instead of water for 10 weeks.The results showed that: Prunus cerasifera Ehrh fruit water extract could significantly reduce the weight of obese mice induced by high-fat diet,reduce the contents of TG and T-CHO in serum and liver,increase the content of SOD in serum,reduce the contents of AKP,AST/GOT,ALT/ GPT in serum,inhibit the growth of adipocytes,and reduce the expression of PPARγ,Fas,SREBP-1c,C/EBP-α,IL-6 and TNF-αgenes in liver.Studies have shown that Prunus cerasifera Ehrh fruit has significant weight loss and lipid-lowering effect on mice fed with high-fat diet,improves the antioxidant activity of the body,has a certain protective effect on liver injury induced by high-fat diet,and inhibits the expression of fat synthesis genes.2.Study on the extraction and component identification of total polyphenols from Prunus cerasifera Ehrh and its antioxidant activity in vitro.The total polyphenols were extracted by alcohol and purified by macroporous resin.The results showed that the content of total polyphenols was 439.17±4.25 mg/g.The main phenolic compounds detected by LC-MS,were,quercetin-7-o-glucoside,quercetin-3-o-arabinoside,quercetin-7-o-arabinoside,ka empferol-3-o-glucoside,kaempferol-7-o-glucoside,kaempferol-7-o-rutinoside,epicatechin3-ocaffeoylquinic acid.The antioxidant activity of total polyphenol extracts detected by different methods was as follows: DPPH radical scavenging rate was 98.95%,total antioxidant capacity of alcohol extracted polyphenols was(0.21±0.01 m M)detected by FRAP method,and(0.48±0.04 m M)detected by ABTS method.3.The intervention effect of polyphenol extract from Prunus cerasifera Ehrh on obesity mice induced by high fat diet.The mice model of obesity induced by high-fat diet was established,and the total polyphenol extract of Prunus cerasifera Ehrh was intervened for 8weeks.The results showed that: Prunus cerasifera Ehrh polyphenols can significantly reduce the weight of obese mice,protect the organs of mice,reduce the contents of TG and T-CHO in serum and liver,reduce the AST/GOT、ALT/GPT in serum,inhibit the enlargement of adipocytes,reduce the accumulation of lipid in liver,and down regulate the genes related to lipid synthesis PPPARγ、C/EBP-α、SREBP-1c 、Fas、ACC、CD36、LPL in liver The expression of IL-6 、 TNF-α 、 IL-1β,CPT-1 、 ACO,Fas 、 C/EBP-α 、 CD36 and the phosphorylation of AMPK were significantly increased.The results showed that the total polyphenols of Prunus cerasifera Ehrh could inhibit the increase of blood lipid by regulating the expression of genes and proteins related to liver fat synthesis and decomposition,and its regulation of lipid metabolism was related to the activation of liver AMPK pathway. |
