Silk,as a significant part of ancient Chinese culture,has promoted the development of world civilization.As early as the Western Han Dynasty,Chinese silk were exported to overseas along the Silk Road and enhanced cultural exchange between Chinese and Western civilization owing to beautiful patterns and rich cultural connotation.However,the origin and transmission of silk has always been an unsolved mystery.Currently,a growing number of scholars have devoted themselves to the exploration of this subject with the unearthing of vast silk relics.The main component of silk is silk fibroin,which is easy to be degraded,leading to the low content of silk protein in unearthed cultural relics.Thus,it's imperative to develop a sensitive method for identifying ancient silk to explore the origin and transmission of silk.In this work,the silk fibroin was used as antigen,and the anti-silk fibroin antibody was obtained by animal immunization.Then the electrochemical immunosensor based on GO/Au nanocomposite and SERS immunosensor based on Au@Ag NRs core-shell nanorods were constructed.After the sensitivity and specificity of the sensor were determined,the immunosensors were used to analyze silk relics.1)A specific polyclonal antibody was prepared to construct a novel electrochemical immunosensor.The GO /Au nanocomposite was synthesized to modify the electrode to improve the electrochemical performance for detection of silk fibroin and the immunosensor revealed a low of detection of 0.58 pg /m L.Additionally,the sensor can effectively quantitatively analyze silk fibroin in cultural relics.Simultaneously,proteomics was also applied to detect the same samples,the consistent detection results were obtained to verify the accuracy and effectiveness of the electrochemical immunosensor.2)A specific monoclonal antibody was prepared to fabricate a SERS immunosensor.Au@Ag NRs core-shell nanorods were synthesized and used in the construction of SERS immunosensor.The immunosensor showed superior performance in detecting silk fibroin,with a detection limit of 7.1 pg/m L.Compared with immunotomography assays,SERS immunosensor had a wider linear detection range and a lower detection limit.In addition,the performance of the sensor was preeminent in the detection of silk relic samples,especially in the qualitative and quantitative analysis of soil samples.In a word,the silk relics identification system based on immunosensor was established,which provides a new methodology for the identification and analysis of silk relics,and also offers a new thought for exploring the origin and transmission of silk.
