ObjectsIn this paper,using the in vitro antioxidant model,UV light damage the skin of mice model,cannabidiol and hemp seed oil on the skin induced by UV light damage of protective effect and mechanism research,and cannabidiol and hemp seed oil of in vitro antioxidant effect,cannabidiol and hemp seed oil as a sun/sunburnt repair products provide preliminary scientific basis.Methods1.DPPH and ABTS free radical scavenging experiments were carried out on cannabidiol and hemp seed oil,and UVB model of Ha Cat cells was constructed to verify the antioxidant activity of cannabidiol and hemp seed oil.2.The mouse UVA+UVB photodamage model was established to evaluate the apparent and skin moisture content of CBD and hemp seed oil to protect the mouse skin photodamage model;3.He staining,Masson staining and Weigert's resorcinol magenta staining were used to observe the effects of cannabidiol and cannabis seed oil on the light-damaged skin tissue structure of mice.4.ELISA,Immunohistochemistry and Western Blot were used to study the protective mechanism of cannabidiol and hemp seed oil against light injury of mouse skin.Results1.The results of in vitro antioxidant efficacy study showed that the IC50 of cannabidiol and hemp seed oil were about 0.75mg/ m L and 70mg/ m L,respectively,and the IC50 of cannabidiol and hemp seed oil were about 1.5mg/ m L and 40mg/ m L,respectively,showing certain antioxidant activity.The validation results of the UVB model of Ha Cat cells showed that cannabidiol had no obvious repair effect on the UVB damaged Ha Cat cells,while hemp seed oil had significant repair effect on the UVB damaged Ha Cat cells.2.The results of UVA+UVB light damage experiment on mouse skin showed that both cannabidol and cannabis seed oil could significantly improve the skin apparent and skin water loss of photoaging mice.The results of H&E staining,collagen staining and elastic fiber staining showed that CBD and hemp seed oil could significantly reduce the abnormal epidermal thickening induced by ultraviolet radiation and inhibit the degradation of collagen and elastic fibers.Meanwhile,skin coating of CBD could not increase the activities of antioxidant enzymes(CAT,SOD and GSH-Px)and the levels of HYP significantly,but significantly reduce the contents of ROS and MDA,and reduce the overexpression of inflammatory factors IL-1?,IL-6 and TNF-?.The expression levels of skin barrier factors FLG,INV and LOR were increased.In addition,cannabidiol can significantly reduce the abnormal secretion of MMP-1 and MMP-3,and inhibit the expression of NF-?B signaling pathway to alleviate the UVA+UVB induced skin light injury.Hemp seed oil could significantly increase the activities of SOD,CAT and GSH-Px and the levels of HYP,while reduce the contents of ROS and MDA.Hemp seed oil can effectively reduce the overexpression of inflammatory cytokines IL-1?,IL-6 and TNF-?,and increase the levels of skin barrier factors FLG,INV and LOR.In addition,hemp seed oil can significantly reduce the abnormal secretion of MMP-1 and MMP-3,and inhibit the expression of NF-?B signaling pathway to alleviate the UVA+UVB induced skin light injury.ConclusionCannabidiol and hemp seed oil have certain antioxidant activity.Cannabidol and hemp seed oil had protective effects on UV-induced skin light injury of mice,reducing the degree of light injury,maintaining the skin moisture content,collagen fiber and elastic fiber,downregulating inflammatory factors and MMPs,and inhibiting the expression of NF-?B signaling pathway.
