| Solid lipid nanoparticles are nano-scale drug-carrying systems made of natural or synthetic solid lipids as framework materials.Using solid lipid nanoparticle embedding technology to encapsulate peptides can ensure better absorption of peptides by the human body,which can effectively improve the utilization rate of the peptide.This topic uses the camel milk polypeptide <3KDa after ultrafiltration as the experimental raw material,adopts the homogenization method to prepare CMP-SLN,optimizes the preparation process of CMP-SLN through single factor and orthogonal experiment design,and analyzes their shape characteristics,Near-infrared analysis,in vitro digestion and stability evaluation.Finally,a camel milk polypeptide nanoparticle oral liquid was developed through single factor and orthogonal experiments.Based on the above experimental results,it is shown that:(1)In this paper,a homogeneous method is used to prepare CMP-SLN.Through single factor and orthogonal experiments,the optimal preparation conditions of CMP-SLN are determined as follows: core/wall 1:2,emulsifier/lipid 1:2.5,water phase/Lipid 16:1.(2)The average particle size of CMP-SLN after embedding is 303 nm,and the polydispersity index(PDI)value is 0.264.The absolute value of the zeta potential is 42.9m V,and the stability is good.Observing the microstructure through transmission electron microscope,CMP-SLN presents independent particles,and the shape is mainly spherical structure.FTIR showed that CMP was successfully embedded in CMP-SLN.(3)In vitro digestion experiments on CMP-SLN.The release rate of CMP-SLN in gastric juice at 0-2 hours was 18.2%,and the release rate in intestinal juice at 3-4 hours was only 44.8%.After passing through the gastrointestinal juice for 4 hours,the inhibition rate of CMP-SLN on DPP4 was 51.2%,and the inhibition rate on α-amylase was 56.9%,which was much higher than that of CMP.The stability study of CMP-SLN showed that it can be stored stably for 30 days at 4°C.At the same time,it has good stability under the temperature conditions of 60℃,70℃ and 80℃.(4)The best process conditions for camel milk polypeptide nanoparticles oral liquid:CMP-SLN addition amount 1%,erythritol addition amount 0.04%,VE addition amount0.01%,citric acid addition amount 0.02%,potassium sorbate addition amount 0.02%,glycerin addition amount 0.2%,sodium carboxymethyl cellulose addition amount 0.02%,xanthan gum addition amount 0.02%.To determine the stability of the oral liquid accelerated test,place it at 37±2℃ for 1~30d.The the final stability rate is 88.33 %,the oral liquid is transparent and uniform in color,and the taste was sweet and sour,suitable for no odor,no precipitation and delamination. |
