Mycotoxin is a toxic secondary metabolite produced by filamentous fungi which has carcinogenic, teratogenic and mutagenic effects, and seriously threatens people’s health.Patulin(PAT)is one of the common mycotoxins, which is very toxic and widely distributed.Zearalenone(ZEN)is one of the most widely polluted mycotoxins in the world, which mainly pollutes sorghum and other grains. Many countries and organizations have formulated clear limit standards for these two mycotoxins residues in food and other products.Because the matrix components of food are complex,it is difficult to detect mycotoxins directly.Solid phase extraction(SPE)is a commonly method for PAT detection, but the traditional adsorption materials couldn’t capture PAT specifically and there application is limmited.Immunoaffinity column method is widely used for zearalenone detection. But its storage condition is harsh, and its detection environment is sensitive to p H,so its application is limited.Molecularly imprinted technology is expected to solve these problems. In this study, the molecularly imprinted polymers(MIPs)of two mycotoxins(patulin and zearalenone)were prepared by using the alternative template technology. And we have established a method to detecting mycotoxins in real samples.Comparing different template molecules, functional monomers, solvents and ratios,we selected 2-indolinone as the template,4-vinylpyridine as the functional monomer, ethyl acetate as the solvent. And the ratio between 2-indolinone and4-vinylpyridine is 1:4.We synthesized the molecular imprinted polymer of patulin by bulk polymerization. Then the molecular imprinted polymer was characterized by SEM,equilibrium adsorption test, adsorption kinetics test and calculation evaluation of imprinted molecule(IF).We found that this molecular imprinted polymer could specifically adsorb patulin. We use the molecularly imprinted polymer of patulin as the adsorption packing to prepare the molecularly imprinted solid-phase extraction column, which could effectively reduce the interference of 5-hydroxymethylfurfural and others in the complex food matrix of apple juice. What’ more, this method could eliminate more than ten kinds of impurity peaks, and the recovery rate reaches 78.2%? 90.1%.And providing a strong support for PAT detection on food safety. We compared the type and proportion of template molecule and functional monomer, and the type of solvent, and finally selected curcumin as the template molecule for zearalenone,MAA as the reagent of functional monomer, acetonitrile(V): methanol(V)= 1:1 as the solvent, and the ratio betwween template molecule curcumin and functional monomer was 1:4.Zearalenone molecular imprint was synthesized by precipitation polymerization trace polymers(MIPs).Its adsorptioncapacity was analyzed by SEM and adsorption experiments, which found that it could specifically adsorb target molecules. The selective extraction of zearalenone toxin from the complex substrate of grain wine was realized by filling the solid-phase extraction column with zearalenone molecularly imprinted polymer, and the content detection of zearalenone was realized by UV spectrophotometer.
