Perfluorinated compounds(PFCs)is a new type of industrial organic environmental pollutants.The food contact materials perfluorooctanoic acid(PFOA)and perfluorooctane sulfonate(PFOS)are typical representatives of perfluorinated compounds.They have strong chemical stability,hydrophobic and oleophobic properties,and can overcome high temperatures and strong oxidants.However,the toxicological studies on them are not perfect,and their food migration and migration patterns are still unclear.In this paper,PFOA was used as the research object,assessment of acute toxicity,genotoxicity and immunotoxicity,and the food migration patterns of PFOA and PFOS was studied.It lays the foundation for the food migration patterns of PFOA and PFOS and the food safety risk assessment,and determine to the population exposure status of the contact materials in foods and its health impact,also the foundation for the development of relevant food safety standards.The results are as follows:1.Studies on the toxicity of PFOA in mice.ICR mice were used as research subjects.According to the toxicological evaluation procedure,the acute oral toxicity,genotoxicity and immunotoxicity of PFOA were studied,which was consistent with the results of foreign studies.PFOA was poisonous,no mutagenic and have immunotoxicity to mice.The acute toxicity of PFOA was studied by Horn method.The results showed that the oral LD50 of female mice was 430 mg/kg,and the oral LD50 of male mice was 369 mg/kg.The acute toxicity was classified as poisoning.The genotoxicity of PFOA was studied by Ames test,mouse bone marrow polychromatic erythrocyte micronucleus test and mouse sperm abnormality test.The results of Ames test showed that in the case of adding and not adding S9,the number of revertant colonies of TA97(a),TA98,TA100,and TA102 strains did not exceed 2 times that of the blank control group,and there was no dose-response relationship;the incidence of micronuclei in the bone marrow polychromatic erythrocytes in three PFOA dosage groups and negative control group was less than 5‰,and there was no significant difference between the dose groups and the negative control group,and there was no dose-response relationship.The results of sperm abnormality test in mice showed that the sperm abnormalities of the three PFOA dose groups and the negative control group were less than 3.4%,and there was no significant difference between the dose groups and the negative control group,and there was no dose-response relationship.The results of three mutagenicity tests indicated that PFOA had no mutagenic effect.The immunotoxicity of PFOA was studied using a carbon particle clearance test,delayed type hypersensitivity test and serum hemolysin assay.The doses of PFOA were set at 20,5,1.5 mg/kg and the negative control group,respectively.The results showed that the phagocytic index?value decreased with the increase of dose in the carbon particle clearance test.The delayed-type allergic reaction test was performed by the method of increasing the thickness of the foot.the difference of the toes gradually decreased with the increase of the dose;the anti-volume in the hemagglutinin test decreased gradually with the increase of dose,indicating that PFOA has an inhibitory effect on the immune function of mice.2.Experimental study on the migration law of PFCs.Taking common paper cups and paper bags as research objects,high performance liquid chromatography-mass spectrometry(HPLC-MS/MS)was used to detect the migration of PFOA and PFOS.There is no established standard for migration at home and abroad.The experiment used WAX solid phase extraction cartridge for enrichment and concentration,C18 column for separation,acetonitrile and water as mobile phase gradient elution,the detection limits and quantitative limits of PFOA were 2×10-3 ng/mL and 7×10-3 ng/mL,respectively;the detection limits and quantitative limits of PFOS were 4×10-3 ng/mL and 1×10-2 ng/mL,respectively.PFOA and PFOS were tested under different operating modes(acid-base neutralization,rotary evaporation)and different environmental conditions(time,temperature)and other factors to different simulants(pure water;4%acetic acid/aqueous solution;the amount of migration in 10%,20%,50%ethanol/aqueous solution,corn oil).Basic data on the migration law of typical PFCs in food contact materials were obtained.Conclusion:The acute toxicity of food contact material PFOA is classified as poisoning,no mutagenicity,and it has certain immunotoxicity in mice in the dose range of1.2520 mg/kg.This indicates that PFOA has certain toxicity,which provides a basis for the dose setting of subchronic toxicity and chronic toxicity studies in the later stage,and also provides data support for the food safety toxicological evaluation of PFOA in China.In the disposable paper cup,the overall migration of PFOA is less than 7.11×10-2 ng/cm2;the overall migration of PFOS is less than 5.38×10-4 ng/cm2;in the packaging paper bag,the overall migration of PFOA is less than 2.7×10-33 ng/cm2;the overall migration of PFOS is less than 1.32×10-3 ng/cm2.At present,there is no exact standard for food migration at home and abroad,providing relevant data on migration and migration laws for future research on PFCs and providing certain data support for the formulation of relevant domestic food standards.
