Pholiota nameko is a nutritious edible fungus,and its polysaccharide has a strong bioactivity.It can effectively improve the immunity,anti-oxidation,anti-tumor,anti-thrombotic,hypoglycemic and lipid lowering effects.These studies combined with the classical chemical analysis,cell experiment and animal experiment to analyze the cell survival rate and telomerase activity by the oxygen peroxide(H2O2)oxidative damage model of the mushroom polysaccharide,and to study the serum interleukins factor and the oxidative stress level of the liver with D-galactose induced aging rat model.This study is aim to explore the antioxidant activity of polysaccharide from the edible mushroom which could improve the more theoretical basis for the polysaccharide.1.The extraction method of hot water extraction was used to extract polysaccharide of Pholiota nameko,and the ratio of extraction liquid,extraction time and extraction temperature were investigated.The response surface test was designed.The result showed that the optimum process conditions for the polysaccharide extraction and alcohol precipitation were:the solid-liquid ratio was 1:25,the extraction time was 3 h,and the extraction temperature was 90?.The highest extraction rate of polysaccharides from the edible mushroom was(5.24±0.156)%.2.Chemical analysis was used to determine the antioxidant activity of the polysaccharide from Pholiota nameko in vitro.The results showed that the polysaccharide of the mushroom could effectively remove the scavenging ability and reducing capacity of DPPH free radical,hydroxyl radical and superoxide anion radical,and there was dose dependence.3.To construct aging model of human embryonic lung fibroblast(MRC-5)induced by H2O2,and to explore the antioxidant effect of polysaccharide from dried mushroom.The results suggested that the optimum concentration of H2O2 inducing cells was 80 mol/L,and the concentration of the polysaccharide was 0.8 mg/mL.Moreover,the mushroom polysaccharide can effectively reduce the damage of cell and enhance telomerase activity of H2O2 treated cells,and can be used as a potential antioxidant.4.The study is aim to establish a rat model of oxidative damage induced by D-galactose,and to explore the antioxidant level in vivo.The results showed that the polysaccharide of mushroom could effectively reduce the content of IL-2 and IL-6 in oxidative damaged rats,and significantly increased the contents of SOD,GSH-Px and T-AOC in the liver of oxidative damaged rats(P<0.01),and significantly reduced the content of MDA(P<0.01).
