[Background]The Lycopene is a natural pigment contained in plants.It is one of carotenoid pigment,as a kind of natural pigment without any side effects of safe,,is one of the most powerful antioxidants currently found in the natural plant.It has many physiological functions,for example anti-cancer,anti-aging,inhibit the growth of malignant tumor,antioxidant and enhance the body immunity and so on.Due to it’s unique physiological role,Lycopene has gradually been recognized by the people.Use of fungal fermentation produce lycopene compared with chemical synthesis method,the activity is maximized.In addition,the method is not only cost saving,but also reduce the pollution and energy consumption and not restricted by season.[Methods]Based on fermentation of Blakeslea trispora,examine cyclase inhibitors and oxygen carrier of metabolic regulation function.Blakeslea trispora was studied by single factor experiment,in the process of fermentation to produce lycopene add tea,theophylline,niacinamide three lycopene cyclase inhibitors’s effect on synthetic quantity of lycopene and bacteria biomass influence.Through the single factor experiment was conducted to study the effect of oxygen carrier in Blakeslea trispora fermentation of beta carotene.Saponification pretreatment on Blakeslea trispora fermentation liquor,using single factor experiment research saponification temperature,saponification time,concentration of KOH three factors on the influence of lycopene extraction yield,and through the orthogonal experiment to optimize the best pretreatment process.Using single factor experiment investigation ultrasonic extracting time,power,material liquid ratio three factors on the influence of lycopene extraction yield,and the ultrasonic extraction process was optimized by using orthogonal method.On the basis of saponification,ultrasonic extraction column chromatography was carried out on the crude extraction and purification,high performance liquid chromatography(HPLC)analysis.The final determination of lycopene on superoxide anion radical(O2-·)and hydroxyl radical(·OH)and DPPH free radical scavenging ability and total antioxidant(T-AOC)in vitro.[Result]Tea as the best cyclase inhibitors,best to join in the 48 h,the best addition amount of 9 g/L,the biosynthesis of lycopene amount can reach 205.49 mg/L.N-hexadecane as oxygen carrier,promote the synthesis of metabolic product beta carotene effect is best,the best time to add 0 h,the optimum adding amount was 6%,138.07 mg/L,increased by 25%than the control group.By saponification method for preprocessing the fermented liquid,the saponification temperature 60℃,time of 48 min,KOH concentration is 0.9 mol/L,can make the lycopene extraction rate reached 43.78%.Through the ultrasonic extraction of lycopene tests showed that the best process conditions for:Ultrasonic power is 300 W,ultrasonic time is 24 min,and solid-liquid ratio is 1:150/(g:mL),under the condition of the extraction yield of 79.2%.This column chromatography purification of lycopene extraction,high performance liquid chromatography(HPLC)analysis was carried on,the final purification of lycopene is 97.48mg/g pure.The results show that the coarse extraction of lycopene of O2-·scavenging ability is very weak,clearance rate lower than 50%.It’s ability to remove·OH,significantly lower than that of VC,IC50 is 300 ug/mL;To the DPPH scavenging ability is a bit weak in beta carotene,IC50is200ug/mL;There is a certain total antioxidant capacity.
