Lonicera tatarica L.and Rosa laxa Ratz.are the unique companion species of the Xinjiang West Tianshan wild fruit forest.It plays an important and irreplaceable role in the geographical climate,biodiversity and ecological defense system of the Yili river basin.In this study,Biological characteristics of reproduction of Lonicera tatarica L.and Rosa laxa Ratz.in the West Tianshan wild fruit forest was studied.the seed dormancy characteristics,cutting propagation characteristics,anatomical characteristics of adventitious roots and tissue culture and rapid propagation characteristics were studied.To provide a scientific basis for the conservation,restoration and sustainable use of the wild Lonicera and Rosa population in the west Tianshan wild fruit forest.The main findings are as follows:(1)By studying the germination characteristics of the seeds of Lonicera tatarica L.and Rosa laxa Ratz.,it was found that the main factor affecting the seed germination of Lonicera tatarica L.was the germination inhibitor present in the seed embryo.The germination of the seeds of Rosa laxa Ratz.was affected by both the seed coat and the germination inhibitors in the embryo.After 60 days stratification and 300 mg/L GA3 treatment,the dormancy of Lonicera tatarica L.seeds was effectively relieved,and the germination rate was 91.10%;For the seeds of Rosa laxa Ratz.,the dormancy of the seeds could be effectively removed by polishing the seed coat,stratification for 80 days and adding 200 mg/L GA3 respectively.The germination rate of the seeds was 64.20%.(2)Through the study on rooting characteristics of the young shoot cutting of Lonicera tatarica L.and Rosa laxa Ratz.,it was found that the survival rate of the young shoot cutting of Lonicera tatarica L.on the river sand substrate was the highest as 56.60%;and the 50 mg/L of IBA:ABT=1:1 mixed solution had the best treatment effect with the survival rate as 62.40%,The average number of roots was 15.10,the average root length was 5.50 cm and the average root diameter as 0.076 cm.On the mixed substrate of river sand+vermiculite+perlite,the cutting survival rate of Rosa laxa Ratz.was the highest,and the survival rate was as high as 65.60%;200 mg/L IBA+ABT=1:1 mixed solution was the best,the survival rate was 69.20%,the average number of roots was 10.40,the average root length was 6.20 cm and the average root diameter was 0.079 cm.(3)Through the anatomical study on rooting morphology of the cutting of Lonicera tatarica L.and Rosa laxa Ratz.,it was found that There was no latent root primordium in cutting of Lonicera tatarica L.and Rosa laxa Ratz.before cutting,and primordium cells were found in cortex after cutting,the adventitious root primordium of Lonicera tatarica L.and Rosa laxa Ratz.cuttings were induced root primordium,and the roots were originated from the cortex.(4)Through the establishment of tissue culture and rapid propagation system of L.and Rosa laxa Ratz.,it was found that MS medium was the most suitable basic medium for the primary culture of stem segments of Lonicera tatarica L.and Rosa laxa Ratz..In the medium MS+6-BA 1.5 mg/L+1BA 0.5 mg/L,the induction rates of Lonicera tatarica L.and Rosa laxa Ratz.were 99.12%and 100%,respectively;The medium suitable for the proliferation of the buds of Lonicera tatarica L.was MS+6-BA 2.0 mg/L+NAA 0.2 mg/L+GA3 1.0 mg/L,and the multiplication coefficient was as high as 5.37;The medium suitable for the proliferation of the buds of Rosa laxa Ratz.was MS+6-BA 1.0 mg/L+NAA 0.2 mg/L+GA3 1.0 mg/L,and the multiplication coefficient was as high as 3.00.The optimum medium for rooting of Lonicera tatarica L.tissue culture seedlings was 1/2MS+NAA 1.5 mg/L+6-BA 0.05 mg/L+AC 1.0 g/L,and the rooting rate was 100%.The optimum medium for rooting of tissue culture seedlings of Rosa laxa Ratz.was 1/2MS+NAA 1.5 mg/L+6-BA 0.1 mg/L+AC 1.0 g/L,and the rooting rate was 66.67%.The survival rates of tissue culture seedlings of Lonicera tatarica L.and Rosa laxa Ratz.transplanted into a single perlite matrix were the highest,The survival rates were 90.00%and 75.00%respectively.
