As the first defense line of human immune defense,natural immunity has attracted more and more attention.Manganese,as an indispensable metal element in the body,plays an important role in natural immunity.Among them,the use of manganese to promote natural immunity is the key point of research.However,manganese has toxicity,the controlled release of manganese is an important problem.In this paper,sodium alginate with good biocompatibility was combined with manganese ions,and manganese-alginate microspheres with different particle sizes were constructed by chemical crosslinking method.The slow-release effect of manganese-alginate microspheres on manganese ions was used to alleviate the toxicity of manganese and give better play to the natural immune function of manganese.The research contents and results are as follows:1.Construction of manganese-alginate microspheres with different sizesManganese-alginate microspheres were constructed by chemical crosslinking method.The effects of sodium alginate concentration,manganese chloride concentration and pH on the particle size of microspheres were discussed,and the preparation conditions of microspheres were optimized.The results showed that the concentration of sodium alginate and manganese chloride were the key factors affecting the particle size,and pH mainly affects the uniformity of particle size distribution.When the concentration of sodium alginate was 0.1 mg/m L,the concentration of manganese chloride was 2mg/m L and pH was 6.2,the minimum particle size of manganese-alginate nano microspheres was 247 nm and the minimum PDI value was 0.2.2.The combination of manganese and sodium alginateIn the process of constructing microspheres,the effects of different anions on the construction of microspheres were studied.The results showed that microspheres could be constructed only by dropping sodium alginate into manganese chloride solution with manganese chloride as crosslinking agent;The constructed microspheres were used to explore the changes of microspheres in specific solutions and the spacing between characteristic peaks in infrared spectra,The results showed that manganese ions play an important role in the structure of microspheres,and the binding mode between manganese ions and sodium alginate was ion crosslinking.The combination of the two methods can better explain the law of the construction of microspheres,and provide a basis for the study of the properties of microspheres.3.Study on the properties of manganese-alginate microspheresThe sustained-release effects of manganese-alginate microspheres with different particle sizes on manganese ions were investigated.The results showed that the microspheres with different particle sizes had sustained-release effect on manganese ions.The release percentage of millimeter scale was 80.6%,while that of micro nano and nano scale are as follows: 86.3%、87.1%;Taking millimeter manganese-alginate as the research object,the water absorption of microspheres was studied.The results showed that manganese-alginate microspheres have strong water absorption,and the swelling ratio was 4800%;The effects of different manganese-alginate microspheres on natural immune properties were further studied by Western blot.The results showed that pH and particle size had a great influence on the immune performance of the microspheres.When the concentration of sodium alginate was 1 mg/m L,the concentration of manganese chloride was 5 mg/m L,and pH was 5.88,the expression of STING protein in the cells treated with micron manganese-alginate microspheres was the highest,indicating that its immune performance was the best.
