Objective:To explore the clinical significance of RANK / RANKL signaling pathway related gene ITGAL in the prognosis of lung adenocarcinoma and its role in immune microenvironment.Methods: the genes of RANK/ RANKL pathway were collected from KEGG database.gene differential expression was analyzed based on TCGA lung adenocarcinoma database.According to the Kaplan Meier curve and clinical characteristics,the final screening gene was identified as ITGAL.Cox risk regression model was used to analyze the overall survival(OS).Then the pathway enrichment of ITGAL gene was analyzed.Finally,CIBERSORT and TIMER modules were used to explore the correlation between ITGAL and tumor immune infiltration.Results: 618 RANK/ RANKL related genes were collected from KEGG database.Bioinformatics analysis of lung adenocarcinoma samples in TCGA database and clinical screening of ITGAL gene.The expression of ITGAL in tumor tissues was significantly increased(P < 0.05),and the high expression of ITGAL was associated with better OS(P< 0.05).Cox regression analysis suggested that ITGAL could be an independent prognostic factor for lung adenocarcinoma(P < 0.05).Gene enrichment pathway showed that the samples with high expression of ITGAL were mainly enriched in immune related signaling pathways.The samples with low expression of ITGAL were mainly enriched in metabolic pathways.Furthermore,CIBERSORT and TIMER database were used to analyze ITGAL,which indicated that ITGAL had a strong correlation with tumor infiltrating immune cells,such as CD8 + T cells,CD4 + T cells,Treg cells,etc,then affected the immune microenvironment of lung adenocarcinoma.Conclusion: ITGAL is a biomarker for the prognosis of lung adenocarcinoma,which can affect the immune invasion of lung adenocarcinoma,and may be a potential predictor of immunotherapy of lung adenocarcinoma in the RANK/RANKL signaling pathway.
