DNA intelligent manufacturing is the most basic layer technology of synthetic biology.The strategic industries relying on the development of DNA intelligent manufacturing,such as DNA information storage,genomic medicine,at least corresponds to the future hundreds of billions to trillions of dollars in the market.High-throughput,high-fidelity,low-cost DNA synthesis technology and its synthetic instruments are the prerequisite and basis for the realization of intelligent DNA manufacturing.Therefore,the development of a high-throughput,low-cost DNA synthesis instrument is of great importance to protect China's biological safety significance.Based on this,this paper analyzes the method of DNA synthesis based on inkjet printing,designs and processes a DNA synthesis chip based on microfluidics,builds a DNA synthesis system based on inkjet printing,and develops the synthesis evaluation and experiment.The main work of the thesis includes the following aspects:(1)Research on DNA synthesis based on inkjet printing technology.This article analyzes the DNA synthesis system based on the specific requirements of DNA synthesis and the technical principles of inkjet printing,studies the surface functionalization of the carrier,the coordination control of the nozzle and the synthesis chip,etc.,thereby discussing the DNA synthesis cycle and process.(2)Structural parameter design and manufacturing of DNA synthesis chips.The DNA synthesis chip is the core device of the DNA synthesis instrument and the reaction carrier for the high-throughput parallel synthesis of DNA.In this paper,the parameters of the liquid channel,the gas channel and the film layer in the synthesis chip are subjected to parameter simulation and simulation experiments in order to determine the corresponding chip structure,and finally the chip manufacturing process and surface functionalization methods were studied.(3)Design of DNA synthesis system based on inkjet printing technology.Build the overall system of inkjet printing for DNA synthesis,including electric drive module,pneumatic drive module,displacement module,visual detection module,etc.,and then design the drive control software,so that the DNA synthesis reagent can be conveyed into the reaction chamber of the synthesis chip according to the customized inkjet print head,and the reagents in the reaction chamber are independently controlled by the microvalve to complete the entire synthesis cycle.(4)Tested and evaluated the DNA synthesis technology and instruments based on inkjet printing.First,the surface functionalization of the synthetic carrier was verified,followed by the analysis experiment of the DNA synthesis product verification method.Due to the low density of DNA synthesis on the surface of the carrier,it could not be analyzed by conventional spectroscopic methods.This paper built a fluorescence in situ hybridization system to evaluate the effect of DNA synthesis.Finally,a DNA synthesis cycle was performed based on manual operations and inkjet printing,respectively,to achieve a complete DNA synthesis process,and to verify the feasibility of the overall system built.
