Research BackgroundFailure of embryo implantation and embryo development is an important factor causing female infertility.CD147 is a highly glycosylated transmembrane glycoprotein,which plays an important role in the proliferation and metastasis of cancer cells,neurodegenerative diseases,and spermatogenesis.CD147 also plays a role in multiple immune responses.The mechanisms of CD147 involved in immunity at the maternal-fetal interface is worth investigating.Low expression of CD147 has been claimed in patients whose repeated failure of CD147 implantation,and infertility has been reported in female mice with CD147 knockout throughout the body.In vitro cytology,CD147 mediated the expression of MMPs family proteins in stromal cells,and MMPs played an important role in the decidualization during implantation.CD147 in utero may play an important role in embryo implantation and embryo development.Therefore,this study adopted the mouse model of conditional uterine CD147 knockout to study the role of CD147 on embryo implantation and the molecular regulation mechanism.PurposesTo investigate the effects of conditional CD147 knockout on various stages of implantation in mice and the mechanism of CD147 regulation in the process of implantation,we used the mouse model of CD147 knockout of uterus.Methods1.PGR-Cre and CD147 Flox mice were mated success to the mouse knockout CD147 in uterus2.The uterus of CD147 mice with conditional uterine knockout at the 5th and 8th day of pregnancy was observed by histomorphology and pathological analysis by HE staining3.PAS staining was used to analyze the effect of conditional knockout uterine CD147 on inflammatory response in decidualized area and the effect of embryo immunity.The effect of Ki67 on the proliferation of stromal cells reflected the effect of conditional CD147 knockout on the fate of stromal cells.4.QPCR was used to detect the levels of IHH,EREG,and other markers of decidualization process in the implantation,as well as the levels of MMPs regulated by CD147 under normal conditions.ResultsThe mouse model of CD147 was successfully constructed by conditional uterine tissue knockout.Western blot and immunofluorescence were used to verify that the knockout effect of CD147 was achieved in uterine luminal epithelium and stromal cells,and there was no effect on ovarian functionAccording to the statistics of the number and weight of implantation sites of uterine tissue on the 5th day of gestation,HE staining and fertility test,it was found that the conditional deletion of CD147 had no effect on the process of embryo adhesion and the fertility of mice in the process of implantation.HE staining,PAS staining and conditional deletion of CD147 had no effect on decidualization and inflammation by counting the number of implantation sites and weight of uterine tissue on the 5th day of gestationConclusionsThis study confirmed that conditional knockout uterine CD147 did not affect the normal fertility of female mice,did not affect the implantation during pregnancy,and had a slight effect on the decidualization process of stromal cells,but was not enough to cause the failure of the entire reproductive event.
