Expression Of Tead1-3in Mouse Uterus During Peri-implantation Period

The implantation of the blastocystinto the maternal uterus is a crucial step in mammalian reproduction, penetration involves invasion of the embryo through the LE and basal lamina into the stroma, to establish a vascular relationship with the mother. At this stage, stromal-cell differentiation into decidual cells (decidualization) is extensive and,like many developmental processes, it involves an intricate succession of genetic and cellular interactions, all of which must be executed within an optimal time frame.In mammals, the four Tead genes are expressed widely, but with distinct patterns, from preimplantation embryos to adult tissues. Most adult tissues express at least one Tead gene。 TEADs are required for cardiogenesis,myogenesis and for the development of neural crest,notochord and trophoectoderm. So far, the expression and regulation of TEAD transcription factors in mouse uterus has not been reported.We study the expression and regulation patterns of Tead1~3in mouse uterus using RT-qPCR and in situ hybridization techniques combined with pregnancy, artificial decidualization, pseudopregnancy, steroid hormone treatment and delayed implantation and activation models. RT-qPCR showed that in early pregnancy the uterus, the expression level of Tead1mRNA was gradually increased and the highest level on day8of pregnancy; the expression level of Tead2mRNA was gradually weakened and the weakest level on day8of pregnancy; the expression level of Tead3mRNA was stable.Tead1~3mRNA were all expressed in the steroid hormone treatment model and reached the highest level on E2treatment model.The signal of Teadl mRNA was detected in the site of implantation in mouse uterus on day5and the decidua of normal pregnant mouse on day6-8of pregnancy, but not in the site of non-implantation; the signal of Tead2mRNA was detected in luminal epithelium and glandular epithelium of uterus on day1~3,there was a weak level expression of Tead2mRNA in decidua of pregnant mouse on day6~8; there was a high level expression of Tead3mRNA in luminal epithelium and glandular epithelium of uterus on day1~4and decidua of pregnant mouse on day6~8.Teadl mRNA and Tead3mRNA were highly expressed in the decidua of the artificial decidualization uterus, there was a weak level expression of Tead2mRNA. Datas from the peseudopregnancy and delayed implantation suggested that the expression of Teadl mRNA and Tead3mRNA were induced by the active blastocyst,but the expression of Tead2mRNA was induced by matrix. The signal of Teadl mRNA was not detected in the steroid hormone treatment model, the signal of Tead2mRNA and Tead3mRNA were detected in E2treatment model. Tead1~3mRNA were relatively highly expressed in the implanted embryos. This research showed that Teadl and Tead3may play an important role in the implantation process and the highly expressed Tead4mRNA and Tead3mRNA in the decidua may be involved in the proliferation and differentiation of decidual cell. Tead2and Tead3may remain the normal fuction of the tissue of epithelium in uterus.
