Fluorescent probes play an important role in the field of life sciences.With the improvement of fluorescence microscopy technology and the development of bioimaging technology,fluorescent probes have become an important tool in the fields of biology and medicine.A large number of studies apply fluorescent probes to specific targeting.The nucleus is the control hub of the cell and contains all the genome of the cell except for a small amount of mitochondrial DNA and plastid DNA in plant cells.In addition,it can also maintain the integrity of the gene and control the activity of the cell by regulating gene expression.Therefore,it is of great significance to develop a probe that can detect the physiological environment of the cell nucleus.In this paper,Hoechst dyes are used as DNA targeting groups and fluorescent donors,and rhodamine and naphthimide derived structures are introduced respectively as recognition groups and fluorescent acceptors for environmental factors,so as to design NO and pH fluorescent probe with nuclear targeting functions.In this paper,Hoechst was used as the DNA targeting group and rhodamine spironolactam was used as the NO indicator,and a nuclear-specific Hoe-Rh-NO ratio probe was prepared.Spectral studies show that the detection limit of the probe reaches 58 nm,is not interfered by substances other than NO,and has a very fast response speed.Bioimaging studies have shown that the probe has good targeting and low toxicity,and can locate and quantify NO in the nucleus.Therefore,Hoe-Rh-NO is a potential probe for detecting NO in the nucleus of living system cells,and it is worthy of evaluation for preclinical applications in the future.Secondly,by combining Hoechst with naphthalimide and using the ICT mechanism,a nuclear-targeted pH probe was obtained.Spectral studies show that the probe has a wide range of pH 10.0-6.5,and the ratio(I550/I430)changes from 0.74 to 1.12.The probe also has good stability.Cell imaging experiments show that the probe has low toxicity and good nuclear localization ability,and can detect the pH of the cell nucleus.
