Objective:In order to provide data support for the prevention and control of diseases transmitted by migratory birds in this region,we analyzed the flora of the faeces of migratory birds in Poyang Lake,found the potential pathogens,and studied the polyphasic taxonomy of strains whose taxonomic status could not be determinedMethods:(1)The faeces of migratory birds in Poyang Lake were mixed with PBS and ground into uniform suspension.The appropriate gradient diluents were spread onto different kinds of culture media under different conditions by the method of isolation and culture.According to the colony morphological characteristics,single colony was selected and purified.Then the 16S r RNA gene sequences were amplified and sequenced.The obtained 16S r RNA gene sequences of the strains were used to blast search against the Gen Bank and Ez Bio Cloud databases.(2)For strains whose taxonomic status cannot be determined,polyphasic taxonomy were analyzed,including colony morphology,cell shape,optimal growth conditions(medium,temperature,p H and salt concentration),biochemical characteristics,phylogenetic analysis and cell chemical composition(fatty acids,polar lipids,respiratory quinones and cell wall compositions).(3)According to the nomenclature rules,new genus and species were proposed.The type strains were deposited to the culture collection centers in different countries,and the corresponding deposit numbers were obtained.Results:(1)In this study,17 untypable strains were isolated from the fecal samples of the migratory birds.They were distributed in 3 phylum,i.e.Actinobacteria,Proteobacteria and Firmicutes.(2)Based on the polyphasic taxonomy,two strains,23H37-10T and 4HC-13,were a novel species of Corynebacterium,named as Corynebacterium anserum sp.nov.,and the type strain was 23H37-10T(=KACC 21672T=GDMCC 1.1737T).Strains23H37-10T and 4HC-13 were isolated from the Greater white-fronted geese(Anser albifrons)in Poyang Lake.Type strain 23H37-10Twas gram-stain-positive,facultatively aerobic,oxidase-negative,catalase-positive,rod-to coccoid-shape,non-motile.Optimal growth occurred at 35–37?,p H 7.0–8.0 and with 0.5–1.5%(w/v)Na Cl.The chromosome size of strain 23H37-10T was 2,208,656 bp(N50,2,208,656 bp;genes,1865)containing 1805 protein-coding genes and the DNA G+C contents was 55.2%.The major fatty acids were C18:1?9c,C18:0 and C16:0.The main polar lipids included diphosphatidylglycerol,phosphatidylglycerol,phosphatidylinositol,phosphatidyl inositol mannosides,two unidentified phospholipids,four unidentified glycolipids and one unidentified lipid.The whole-cell sugar comprised arabinose.(3)Based on the polyphasic taxonomy,strains C64T and C62,were a novel genus of family Actinomycetaceae,named as Nanchangia anserum gen.nov.,sp.nov.,and the type strain was C64T(=CGMCC 1.18409T=GDMCC 1.1969T=KCTC49511T=KACC 22143T).Strains C64T and C62 were isolated from the Greater white-fronted geese(Anser albifrons)in Poyang Lake.Type strain C64T was gram-stain-positive,anaerobic,oxidase-negative,catalase-positive,rod-shaped,non-motile.Optimal growth occurred at 37?,p H7.0 and with 0.5%(w/v)Na Cl.The chromosome size of strain 23H37-10T was 1,907,497 bp containing 1789 genes in total and 1593 genes were protein-coding genes.The DNA G+C contents of the type strain was 66.0%.The major fatty acids were C18:1?9?and C16:0.The main polar lipids included phosphatidylinositol,phosphatidylglycerol,phosphatidylcholine,diacylglycerol,triacylglycerol,and cardiolipin.The whole-cell sugar comprised galactose,arabinose,and glucose.Conclusion:There are abundant gut bacteria in the faeces of migratory birds in Poyang Lake,and there are many new(genus)species of bacteria that are not yet recognized.This study identified and named the new species of Corynebacterium anserum and the new genus of Actinomycetaceae from Anser albifrons,which enriched the resources of intestinal related bacteria of migratory birds,but its biological significance still needs further study.
