There are abundant microbial resources in the environment of Tengchong Hot Spring,and researchers have continuously discovered many new species,new thermostable enzymes and secondary metabolites from Tengchong.Hot Spring.Therefore,the mining of microbial resources in Tengchong high-temperature hot springs has been a research hotspot in recent years.Based on the current reports on the microbial-related biodiversity of Yunnan hot springs,most of them are concentrated in Tengchong Hot Springs Rehai,Jinze,etc.However,there are few studies on excavation reports on the microbial-related biodiversity of Tengchong hot springs,Gumingquan and Hamazui,and pure culture isolation and identification.This study is based on two hot springs in Tengchong,Guming Spring and Hamazui.Through high-throughput sequencing analysis and pure culture methods,the bacterial diversity analysis and the isolation and identification of some cultivable bacteria in the two major hot springs were carried out,and the main results are as follows:(1)Through high-throughput sequencing analysis,a total of 67 phyla,174 classes,380 orders,600 families,and 1038 genera were detected in 9 samples collected from Hamazui and Gumingquan(data after deduplication).At the same time,there are significant differences in 57 genera and 95 genera of bacteria at the sampling points on the Hamazui slope and on the side of the road.There are a total of 55 genera of bacteria in Guming Spring sampling sites with significant differences,indicating that the bacterial communities in the two major hot springs have high abundance;(2)Through the analysis of the related functions of the Hamazui and Guming spring bacterial community and the abundance of metabolic pathways,the results show: the biosynthesis of secondary metabolites of microorganisms in the two major hot springs,the transportation and metabolism of coenzymes,lipids and carbohydrates,energy production and conversion,amino acid transport,metabolism and other related functions have high abundance,and unknown functions(such as unknown secondary metabolite genes and related synthesis pathways)are relatively abundant.The above results show that the two hot springs have the significance and value of in-depth exploration in the discovery of new microbial strains,the development of secondary metabolites such as antibiotics,and the search for high-temperature and high-efficiency unknown enzymes;(3)The hot spring microorganisms in the Guming spring and the Hamazui have been isolated and cultured by the dilution coating plate method.The results showed that a total of 164 strains of bacteria were obtained in pure culture.These bacteria were distributed in 4 phyla,6 classes,18 orders,24 families and 35 genera,including35 potential new bacterial species.Statistical analysis of the separation effect of each separation medium showed that the separation effect of nitrogen fixation medium and T3 medium was the best,followed by T5 medium and denitrification medium,and Gao's medium was the worst;(4)A new strain of YIM B00624 was sequenced and annotated,and the effective data was about 1 G.The total length of the complete genome was 8.771 M,and the GC content was 53.55%.Genomic analysis results show that the bacteria YIM B00624 contains very rich protein domains,abundant transmembrane transport mechanisms,protein transport mechanisms,energy conversion mechanisms,and metal ion binding mechanisms.At the same time,through the annotation of metabolic pathways,it is found that there are related mechanisms of signal transduction and membrane transport-related environmental processing.It shows that in the hot spring environment,hot spring microorganisms continuously secrete secondary metabolites to inhibit or kill the growth of other bacteria,and obtain external nutrients through membrane transport for their own growth.This is the continuous evolution of microorganisms that have adapted to the hot spring environment for a long time.
