Haematococcus Pluvialis is a kind of monocelled freshwater green algae which is widely found in nature,it will accumulate staxanthin and become haematocysts under environmental stress,especially under high light stress.Astaxanthin is a pigment with high economic value,and the extraction of astaxanthin from Haematococcus Pluvialis is the most commonly used method of astaxanthin production.Because it is extremely sensitive to changes in light intensity,it is difficult to directly improve the biomass of Haematococcus Pluvialis by directly improving the light intensity.This paper explores the growth of Haematococcus Pluvialis by changing the culture light wavelength.In addition,the photosynthetic efficiency of photosynthetic organisms is closely related to light,researching on photosynthetic characteristics of Haematococcus has certain theoretical value for photosynthetic efficiency mechanisms of photosynthetic organisms.This paper studies the effects of different monochromatic light to the growth of Haematococcus and ROS accumulation for researching its photosynthetic efficiency mechanisms,by fluorescence staining and chlorophyll fluorescence parameters,and hopes that such changes can be microinterpreted by fluorescence quantitative PCR.Results are follows:(1)A simple monochromatic light irradiance measuring device is made by using the simple connection between the small solar panel and the voltmeter phase which supplied by a general mobile power supply.According to the curve fitting equation L=1.524*e exp1.525V(e is the base of the natural logarithm),we can easily measure the radiation strength of any monochromatic light by the measurement of voltage.(2)The fluorescence microscopy and chlorophyll fluorescence parameters show that the light culture of 460 nm wavelength is most conducive to the growth of Haematococcus.(3)By contrast of the fluorescence parameters of different chlorophyll,it shows that 460 nm is more beneficial to improve its photocooperation and electron transfer efficiency,while 550 nm improves its antioxidant stress resistance.(4)The expression of rbc L,psa B,psb A,SOD,APX genes in 550 nm,425nm culture was higher than 460 nm,660nm and fluorescent culture.In this experiment,a simple monochromatic light irradiance measuring device is successfully made.With this simple device,the irradiance of monochromatic light can be measured and it is equal to irradiance of sunlight of certain illuminance.Using this device can reduce the cost of light intensity measurement in the study of the influence of monochromatic light on plants.Experiments found that blue light culture can have high growth rate and photosynthesis rate and facilitate the split propagation of Haematococcus,which has of practical significance to improving the cultivation efficiency of Haematococcus.The obvious differences in photosynthesis efficiency between green light and blue light and red light culture and the changes in photosynthesis related gene expression reveal that the changes of photosynthetic efficiency may be affected by various factors.
